摘要
目的研究抗过敏药枸地氯雷他定对卵蛋白致敏小鼠脾淋巴细胞表达炎症因子的影响,探讨其抗过敏性炎症活性。方法小鼠腹腔注射卵蛋白联合氢氧化铝胶体构建致敏动物模型,无菌分离小鼠脾淋巴细胞,以不同浓度药物进行阻断处理,酶联免疫吸附法(ELISA)检测培养细胞上清液中白三烯(LTB4)、前列腺素E2(PGE2)、白介素4(IL-4)、白介素5(IL-5)及肿瘤坏死因子(TNF-α)的水平。结果枸地氯雷他定不同浓度可明显抑制致敏淋巴细胞释放PGE2、TNF-α、IL-4、IL-5等炎症因子,且具有浓度依赖性。结论枸地氯雷他定有显著的抗过敏性炎症活性。
Objective To investigate the effect of Desloratadine Citrate Disodium on the release of several inflammatory mediators such as leukotfiene B4( LTB4),interleukin4( IL-4),interleukin5( IL-5),prostaglandin E2( PGE2) and tumor necrosis factor α( TNF-α)from spleen lymphocytes of allergic mice challenged by ovalbumin( OVA). Methods OVA-sensitized mice were used as animal model,spleen lymphocytes isolated from mice were treated with desloratadine citrate disodium,ebastine,levocetirizine hydrochloride and dexamethasone,followed by incubation with OVA. These cells without pretreatment with drugs served as the control. Enzyme 1inked immunosorbent assay( ELISA) was applied to measure the level of these inflammatory mediators. Results Desloratadine citrate disodium significantly inhibited the release of LTB4,IL-4,IL-5,PGE2 and TNF-α in a dose dependent manner. Conclusion Desloratadine citrate disodium has a potent antagonistic effect on the allergic-inflammation.
