摘要
为了研究脱皮激素受体在拟黑多刺蚁发育中的功能,本项目采用逆转录PCR方法从拟黑多刺蚁(Polyrhachis vicina Roger)中克隆到脱皮激素受体编码基因PvEcR全长序列,对其编码的氨基酸序列进行生物信息学分析。从拟黑多刺蚁中克隆到脱皮激素受体蛋白编码基因1 737bp的全长序列,该序列编码578个氨基酸,预测的蛋白分子量大小为63.098×103,理论等电点为7.41。NCBI蛋白质数据库中进行Blast搜索表明,拟黑多刺蚁脱皮激素受体蛋白PvEcR存在至少6类保守结构区域,主要为DNA结合位点、配体结合位点和激活因子识别位点。PvEcR蛋白序列磷酸化位点预测共揭示48个可能的磷酸化位点。PvEcR与其他昆虫脱皮激素受体蛋白在氨基酸序列上同源性高达70%以上。基于脱皮激素受体蛋白氨基酸序列构建的系统发育树表明,几乎所有蚁类脱皮激素受体形成一个大分支,其中拟黑多刺蚁与佛罗里达弓背蚁表现出最近的亲缘关系。
To reveal the function of ecdysone receptor in the development of Polyrhachis vicina,full-length coding region sequence of Polyrhachis vicina ecdysone receptor with the length of 1 737 bp was cloned using reverse transcription-Polymerase Chain Reaction(RT-PCR)in this study.This sequence encodes a protein of 578 amino acids with molecular weight of 63.098×103 and the theoretical isoelectric point of 7.41.Blast search against protein database in NCBI showed that Polyrhachis vicinaecdysone receptor(PvEcR)contains at least 6highly conserved domains,mainly being DNA binding sites,ligand binding sites,and activator recognition sites.Phosphorylation site prediction of PvEcR revealed 48 putative phosphorylation sites that might play important roles in hormone signal pathway.PvEcR showed no less than 70% similarity with ecdysone receptor proteins from other insect species.Phylogenetic tree that was constructed based on amino acid sequences of these insect ecdysone receptor proteins suggested that almost all insect ecdysone receptors form a large subfamily,among which Polyrhachis vicina ecdysone receptor shows the closest phylogenetic relationship with Camponotus floridanus ecdysone receptor.
