摘要
目的 :研究支架术后不同时间局部食管组织中相关细胞因子的表达及其与局部再狭窄形成的关系。 方法 :采取“自体阔筋膜移植固定法”置入“Z”型自扩张金属食管支架 ,于术后 1、2、4、8周分批处死动物 ,取出置架部位的再狭窄食管组织 ,采用免疫组化 (ABC法 )检测支架术后食管周组织中 TNF-α、TGF-β1 、PDGF、ET1 的表达及其变化规律。结果 :支架术后 1周组织即开始表达 TNF- α、ET1 ;术后 2周 TNF- α、TGF- β1 、PDGF、ET1 表达均明显增强 ;术后 4周仍有少量 TGF- β1 表达 ;术后 8周所有细胞因子均极少表达。着色阳性的细胞主要是巨噬细胞、T淋巴细胞及肥大细胞 ,ET1 主要由血管内皮细胞表达。 结论 :TNF- α、TGF- β1 、PDGF、ET1 作为炎性细胞的表达产物 ,对纤维化及再狭窄的形成具有重要调控作用 ,与支架术后再狭窄的形成有密切关系。
Objective: To study the expression of related cytokines and its relation with re stenosis after stenting.Methods:Healthy adult dogs were used as experimental subjects.Esophageal stent was placed by the method of 'autogenous broad fascia transplantation and fixation'. The dogs were killed at the end of 1,2,4,8 weeks,and the esophageal re stenosis tissue with stent were taken as sample.The expression of TNF α,TGF β 1,PDGF and ET 1 in esophageal tissue after stenting were studied by immunohistochemical method(ABC). Results:One week after stenting esophageal tissue began to express TNF α and ET 1,2 weeks after stenting the expression of TNF α,TGF β 1,PDGF,ET 1 were increased,at week 4 there was still a little expression of TGF β 1,all cytokines almost disappeared at week 8. The positive cells were mainly Mφ,T cell and mast cell,ET 1 expressed mainly in vascular endothelial cells of blood vessel. Conclusion:TNF α,TGF β 1,PDGF,ET 1 produced by inflammatory cells may act as important regulators in fibrosis and restenosis.
