摘要
目的 建立非肌肉浸润性膀胱癌裸鼠皮下移植瘤模型.方法 采用中国人膀胱癌BIU-87细胞系,并应用Matrigel基质胶作为细胞悬浮剂将癌细胞接种于裸鼠腋窝皮下,记录成瘤时间,计算成瘤百分率,测量肿瘤长径、短径、重量,绘制肿瘤生长曲线,并进行病理学检查.结果 裸鼠皮下移植瘤模型的成瘤时间约为6~7d,成瘤率达到了100%,皮下接种28d后移植瘤平均长径为(19.58±1.85) mm,平均短径为(14.18±1.59)mm,平均体积为(2098.13±556.88)mm3,平均瘤重为(0.92±0.27)g.移植瘤HE染色显示,癌细胞的大小不一,排列紊乱,极性消失,核分裂像多见,细胞核染色深.结论应用Matrigel基质胶作为癌细胞的悬浮剂成功建立了裸鼠皮下移植瘤模型,为后续进行非肌肉浸润性膀胱癌的科学实验研究奠定了基础.
Objectives To establish non-muscle invasive bladder carcinoma xenograft model in nude mice.Methods We injected the human bladder carcinoma cell line BIU-87 suspended in Matrigel into nude mice,recorded the incubation period and calculated the forming tumor percentage,measured long diameter,short diameter and weight of tumors,and then carry out the pathological identification of tumors.Results All nude mice formed transplanted tumors.The average time of their formation was about 6 ~ 7 days.The average long diameter and short diameter of these tumors were about (19.58 ± 1.85) mm and (14.18 ± 1.59) mm,the average tumor volume were (2098.13 ± 556.88) mm3 and the average tumor weight of (0.92 ± 0.27) g.Pathological identification showed that cancer cells arranged is similar to transitional cell,but shows greater variability in cell size,shape,and greater loss of polarity.Conclusions Applying Matrigel as a suspending agent successfully established superficial bladder cancer xenograft model in nude mice,and lay a foundation for further investigation on non-invasive bladder carcinoma.
