摘要
目的探讨大肠癌细胞中人类白细胞分化抗原133(CD133)与八聚体结合转录因子4(OCT4)的表达情况及其相关性。方法采用磁珠细胞分离技术分选出人结肠癌细胞系的CD133+细胞和CD133-细胞,检测两种细胞中OCT4的表达。分别改变CD133+和CD133-细胞中OCT4的表达,观察其对CD133的表达的影响。采用肿瘤球形成实验检验肿瘤细胞干性生物学行为的改变。结果 CD133+细胞中OCT4呈现高表达,CD133-细胞中OCT4基本无表达。CD133+细胞对5-氟尿嘧啶(5-FU)高度抵抗,并且更容易在无血清培养基中形成肿瘤球。干扰OCT4的表达可以降低CD133+细胞中CD133的表达水平,增加对5-FU的敏感性,并且减少肿瘤球形成能力。过表达OCT4增加CD133-细胞CD133的表达水平,降低对5-FU的敏感性,并且减少肿瘤球形成能力。结论 OCT4与CD133在大肠癌细胞中均有表达,OCT4对CD133有直接调控作用。
Objective To explore the expressions of human leukocyte differentiation antigen 133( CD133)and octamer binding transcription factor 4( OCT4) in colon cancer cells and their correlation. Methods CD133+cells and CD133-cells were separated by immunomagnetic separation technology. Their OCT4 expressions were detected and changed,and the effect of OCT4 expressions on CD133 expression was observed. Tumor sphere formation assay was employed to test the cancer stem cell phenotype. Results CD133+cells showed a high expression of OCT4,and CD133-cells showed no expression of OCT4. CD133+cells were of high resistance to 5-fluorouracil( 5-FU),and were easier to form spheres in the serum-free medium. Inhibiting the OCT4 expression reduced the CD133 expression in CD133+cells,increased the 5-FU sensitivity,and reduced the tumor sphere formation ability. Over-expression of OCT4 increased CD133 expression in CD133- cells,decreased 5-FU resistance,and reduced tumor sphere formation ability.Conclusion Both OCT4 and CD133 are expressed in colon cancer cells,OCT4 may play a role on regulating CD133 directly.
