摘要
目的测试番荔枝内酯单体Bullatacin,研究其对ER(-)人乳腺癌细胞MDA-MB-231增殖的影响,为抗乳腺癌新药开发提供实验依据。方法 MTT法观察Bullatacin体外对MDA-MB-231细胞增殖的抑制作用,并筛选出有效剂量和作用时间;流式细胞仪检测其对细胞周期的影响。结果 Bullacatin对MDA-MB-231细胞增殖抑制有剂量和时间依赖性;能使G1期的细胞比率减少,S期的细胞比率增加,细胞周期明显阻滞在S期。结论 Bullatacin可以抑制MDA-MB-231细胞的增殖,使细胞周期阻滞在S期。
OBJECTIVE To investigate the effects of Bullatacin on proliferation of breast cancer cell line MDA-MB-231.METHODS MTT assay was used to observe the in vitro inhibitory effect of Bullatacin on MDA-MB-231cells and to screen effective dose and active time;Flow cytometry was applied to explore its effects on cell cycle.RESULTS The inhibitory effects of Bullacatin on the proliferation of breast cancer cell line MDA-MB-231were related to the dose and time;it can reduce the cell ratio of G1 phase,increase the cell ratio in S phase,and obviously maintain the cell cycle in S phase.CONCLUSION Bullatacin may inhibit the proliferation of breast cancer line MDA-MB-231and prevent the growth of cancer cells staying in S phase.
