羟基喜树碱聚氰基丙烯酸正丁酯纳米囊制备工艺研究 被引量:2

摘要 目的制备羟基喜树碱聚氰基丙烯酸正丁酯纳米囊(HCPT-PBCA-NPs),研究制备工艺并优化处方。方法采用微乳化聚合法制备HCPT-PBCA-NPs,以粒径、多分散系数和包封率为指标,通过单因素考察处方和工艺参数,采用Box-Behnken实验设计优化HCPT-PBCA-NPs制备工艺并考察其理化性质。结果按照优化处方和工艺条件,制得纳米囊平均粒径为(92.7±0.6)nm,多分散系数为0.118±0.014,包封率为(94.24±1.05)%,形态圆整,具有缓释作用。结论经过优化筛选的处方和制备工艺制备的纳米囊具有较好的理化性质,缓释效果显著。 OBJECTIVE To prepare hydroxycamptothecin loaded Poly(butyl cyanoacrylate)nanocapsules(HCPT-PBCANPs),and study on the preparing technology and optimize the formulation.METHODS HCPT-PBCA-NPs was prepared by micro-emulsion polymerization,the preparing technology was optimized by single factor test and Box-Behnken design with size distribution,polydispersion index and encapsulation efficient was set as the test standard.In addition,its characteristics were investigated.RESULTS Under the optimum condition,the size distribution of HCPT-PBCA-NPs was(92.7±0.6)nm,polydispension index was(0.118±0.014),and encapsulation efficient was(94.24±1.05)%.Meantime,the HCPT-PBCANPs nanocapsules were smooth and spherical was observed by TEM and AFM with sustained-release.CONCLUSION The optimal HCPT-PBCA-NPs show good characterizatics and sustained-release in vitro.
