摘要
目的通过检测大肠癌组织及相应癌旁组织中的C-Jun结合蛋白1(c-Jun activation domain binding protein 1,JAB1)和β-连环素(β-catenin)的表达,探讨JAB1和β-catenin在大肠癌组织中的表达与其生物学行为之间的关系。方法 (1)应用免疫组化学方法(SP法)检测94例大肠癌及相应癌旁组织中JAB1和β-catenin的表达情况。(2)分析JAB1和β-catenin的表达与大肠癌的发生、发展、分化程度、临床分期及淋巴结转移等之间的关系。结果 (1)JAB1在大肠癌组织中的表达率明显高于癌旁组织(χ2=86.686,P<0.001),JAB1在低分化型大肠癌中的阳性表达率为94.3%,高于中、高分化型中的79.7%(χ2=5.634,P=0.018);在淋巴结转移阳性的病例中表达率为94.6%,高于淋巴结转移阴性的病例中的73.7%(χ2=8.344,P=0.004);在Dukes分期C期+D期中的阳性表达率为93.2%,在A期+B期中阳性表达率为70.0%,差异具有统计学意义(χ2=8.124,P=0.004);JAB1的表达与患者性别、年龄、肿瘤大小无明显统计学意义(P>0.05);(2)在94例大肠癌组织中β-catenin异常表达数为61例(64.9%),癌旁组织异常表达数为26例(27.7%),二者差别有明显统计学意义(χ2=26.209,P<0.001),β-catenin在低分化型大肠癌中的异常表达率为91.4%,高于中、高分化型中的72.9%(χ2=4.686,P=0.030);在淋巴结转移阳性的病例中异常表达率为98.2%,高于淋巴结转移阴性的病例的76.3%(P=0.001);在Dukes分期C期+D期中的异常表达率为90.9%,在A期+B期中异常表达率为64.0%,差异具有统计学意义(χ2=9.454,P=0.002)。β-catenin的表达与患者性别、年龄、肿瘤大小无明显统计学意义(P>0.05)。结论 (1)JAB1、β-catenin参与大肠癌的发生及发展;(2)JAB1可能会成为大肠癌新的肿瘤标记物,提高其诊断率。
Objective To investigate the expression of JAB 1 andβ-catenin in colorectal carcinoma and its biological behavior relation-ships by detecting colorectal cancer and corresponding paracancer tissue .Methods We used immunohistochemistry ( SP method ) to detect the expression of JAB1 andβ-catenin in 94 cases of colorectal cancer and corresponding paracancer tissues .The relationship be-tween JAB1 and β-catenin expression and development ,degree of differentiation ,clinical stage ,lymph node metastasis and prognosis of the colorectal cancer were analyzed .Results The expression of JAB1 in colorectal cancer rate was significantly higher than correspond-ing paracancer cancer tissues (χ2 =86.686,P&lt;0.001).The positive expression rate of JAB1 in poorly differentiated colorectal cancer (94.3 %) was higher than that in highly differentiated colorectal cancer (79.7%%) (χ2 =5.634,P =0.018).JAB1 expression in lymph node positive cases (94.6 %) was higher than that in lymph node negative cases (73.7%) (χ2 =8.344,P=0.004).The positive expression rate in Dukes stage C +D period (93.2%) was significant different from that in the A +B period (70.0%) (χ2=8.124,P=0.004);The correlation between the expression of JAB1 and sex,age,tumor size was not statistically significant (P〉0.05).In 94 cases colorectal cancer ,the number of abnormal expression of β-catenin was 61 cases(64.9%),significantly different from that of corresponding paracancer tissues ,26 cases(27.7%) (χ2 =26.209,P&lt;0.001).The abnormal expression rate of β-catenin in poorly differentiated colorectal cancer (91.4 %) was higher than that in well differentiated colorectal cancer (72.9%) (χ2 =4.686,P=0.030).The abnormal expression rate of lymph node positive cases (98.2%) was higher than that of lymph node negative cases (76.3%)(P=0.001).The abnormal expression rate in Dukes stage C +D period (90.9% ) was signifantly different from that in stage A +B period (64.0%)(χ2 =9.454,P=0.002).The correlation between the expression of β-catenin and sex,age, tumor size was not statistically significant (P〉0.05).Conclusions JAB1,β-catenin were involved in the incidence and development of colorectal cancer .JAB1 could be a new tumor marker for colorectal cancer to improve the rate of diagnosis .
