响应面分析法优化湿地松松针中酪氨酸酶抑制剂提取工艺 被引量:2

摘要 在料液比、乙醇浓度、提取温度和提取时间这4个单因素试验的基础上,利用响应面分析法优化湿地松松针酪氨酸酶抑制剂的提取工艺。结果表明,最佳提取工艺为:料液比1∶18,乙醇浓度55%,浸提温度85℃;此条件下,提取液对酪氨酸酶的抑制率达70.67%;湿地松松针的粗提物的得率为21.43%,对酪氨酸酶抑制率的IC50为0.91 mg/mL。采用响应面分析法可以对湿地松松针酪氨酸酶抑制剂提取工艺进行优化。 On the basis of single factor experiments, the method of response surface analysis was adopted to approach an optimal extraction process of tyrosinase inhibitor. Results showed: The optimised extraction conditions for the tyrosinase inhibitor were obtained with solid/liquid ratio, temperature, the concentration of ethanol as 1/18, 85 ℃, 55%, respectively. Under these conditions, the tyrosinase inhibition rate of the extracted liquid (5 mg/mL) was up to 70.67%, With the extraction value of 21.43%, the extracts had significant tyrosinase inhibitory activity with the IC^0 of 0.9l mg/mL. Conclusion is that response surface method is feasible for the optimization of the extraction process for tyrosinase inhibitor from pine needles of P/nus elliottii, and the optimized process is rehable.
