摘要
目的研究血必净对重症急性胰腺炎(SAP)大鼠TLR9信号通路调控肠黏膜炎症反应机制的影响。方法选择30只SD大鼠作为研究对象,以数字随机法分成空白组、对照组和治疗组,每组10只。对照组和治疗组以牛黄胆酸钠建模,空白组则以生理盐水建模。对治疗组注射血必净药物。观察并对比3组定量检测指标的表达水平,以及各组血清淀粉酶、TNF-a、IL-6、IL-10、二胺氧化酶(DAO)和D-乳酸水平情况。结果治疗组的TLR9mRNA、TLR9蛋白及NF-κB水平在治疗后相对于对照组均有下降,而对照组相对于空白组TLR9mRNA、TLR9蛋白及NF-κB水平显著上升,且两两对比,差异均有统计学意义(P均<0.05)。治疗组的血清淀粉酶、TNF-α、IL-6、DAO及D-乳酸水平与对照组相比均显著下降,IL-10水平显著上升,对照组的血清淀粉酶、TNF-α、IL-6、DAO及D-乳酸水平与空白组相比,显著上升,IL-10水平显著下降,差异均有统计学意义(P均<0.05)。结论 TLR9介导的有关信号通路可能参与到SAP发病过程。而血必净注射液可降低血清炎症介质的水平,对SAP大鼠的肠黏膜屏障发生的损伤有一定保护作用。
Objective To study impact of Xuebijing injection on mechanism of TLR9 signaling pathway regulating intestinal mucosal inflammation reaction in Severe acute pancreatitis(SAP) rats.Methods 30 SD rats in hospital were selected as research objects, and were divided into blank group, control group and treatment group by digital random methods. Modeling of control group and treatment group was founded with sodium taurocholate, and control group was founded with saline. Treatment group was injected with Xuebijing drugs. Quantitative detection of the expression levels of three groups’ indicators, as well as the serum amylase, TNF-α, IL-6, IL-10, DAO and D- lactate levels circumstances were observed and compared .Results The treatment group’s TLR9mRNA, TLR9 and NF-κB protein levels after treatment were decreased compared to the control group, while TLR9mRNA, TLR9 and NF-κB protein levels of control group, compared to blank group, were significantly increased, and with pairwise comparison. The differences were statistically significant (P〈0.05). The treatment group’s serum amylase, TNF-α, IL-6, DAO and D- lactate levels compared with control group were significantly decreased, with a significant increase in IL-10 levels. Serum amylase, TNF-α, IL- 6, DAO and D- lactate level in the control group, compared to blank group, had a significant increase. And IL-10 levels were significantly decreased, and differences were statistically significant(P〈0.05).Conclusion TLR9-mediated signaling pathway may be involved in the pathogenesis of SAP. Xuebijing injection can reduce the serum levels of inflammatory mediators, and has a protective effect in damage of SAP mucosal barrier occurring in rats.
