摘要
目的研究猪血红蛋白的酶解工艺。方法用组合酶Ⅰ(蛋白酶F与蛋白酶P)和组合酶Ⅱ(蛋白酶F与蛋白酶N)同步酶解猪血红蛋白。结果确定最佳工艺参数,组合酶Ⅰ的最佳酶解条件为:酶添加量0.2%,温度50℃,料水比1:1.25,pH 7.5,水解时间20 h;组合酶Ⅱ的最佳酶解条件为:酶添加量0.24%,温度50℃,料水比1:1.75,pH 8.0,酶解时间20h。此工艺条件下,组合酶Ⅰ和组合酶Ⅱ三氯乙酸可溶性氮含量(TCA-SN指数)分别为71.72%和87.82%,两种组合酶的蛋白质回收率均达90%。结论组合酶Ⅱ较适于工业生产。
Objective To study the enzymolysis technology of porcine hemoglobin.Methods Porcine hemoglobin was synchronously hydrolyzed by the mixed enzyme Ⅰ (Protease F and Protease P) and the mixed enzyme Ⅱ (Protease F and Protease N).Results The optimum conditions for the mixed enzyme Ⅰ were as follows:enzymatic addition 0.2 %,enzymolysis temperature 50 ℃,ratio of material to water of 1∶1.25,pH 7.5,enzymolysis time 20 hours.The optimum conditions for the mixed enzyme Ⅱ were as follows:enzymatic addition 0.24 %,enzymolysis temperature 50 ℃,ratio of material to water of 1:1.75,pH 8.0,enzymolysis time 20 hours.Under those conditions,the protein recovery rate of two kinds of the mixed enzyme both reached 90% and the trichloroacetic acid-soluble nitrogen (TCA-SN) contents were 71.72 % and 87.82 %,respectively.Conclusion The mixed enzyme Ⅱ can be suitable for the industrial production.
