摘要
目的建立同时测定多种类海产品(鱼类、软体类、甲壳类等)中37种脂肪酸含量的快速、多组分检测技术,同时对采集的海产品进行监测。方法脂肪酸经Folch法提取,氯乙酰-甲醇法甲酯化,石油醚提取,气相色谱FID检测器测定,以十九酸甲酯为内标定量计算。色谱柱:DB-225毛细管柱(20.0 m×0.10 mm×0.10μm);升温程序:起始温度35℃,保持0.5 min,以25℃/min升至195℃,以3℃/min升至205℃,以8℃/min升至230℃保持5 min;载气流速(氢气):1 ml/min。结果 37种脂肪酸的检测限范围:3.55 mg/100 g^124.66 mg/100 g,回收率范围:66.85%~131.74%,相对标准偏差(RSD):7.39%~9.98%。对采集的5种30份海产品样本检测,发现小黄鱼体内ω-3不饱和脂肪酸含量高于其它种类的海产品。结论该方法操作简便、耗时短、线性良好,能满足不同种类海产品类中37种脂肪酸的检测,为海产品营养成分监测与评估提供技术支持。
Objective To develop a simple and rapid gas chromatography method for simultaneous determination of 37 fatty acids in seafood and to monitor the seafoods from Zhoushan Island. Methods Samples were extracted with Folch method,followed by methyl esterification with acetyl chlorde- MeOH,then extracted by petroleum ether,analyzed by GC with FID as detector and methyl nonadecanoate as the internal standard for quantitation calculation. The chromatographic column was DB- 225( 20.0 m ×0. 10 mm ×0. 10 μm),and the temperature program was set with the oven temperature at 35 ℃ for 0. 5 min,ramped to 195 ℃ at 25 ℃ /min,ramped to 205 ℃ at 3 ℃ /min,ramped to 230 ℃ at 8 ℃ /min and kept for 10 min at 230 ℃,the flow rate of carrier gas was 1 ml /min. Results The detection limits of 37 fatty acids in seafood ranged from 3. 55 mg /100 g to 124. 66 mg /100 g. The average recoveries ranged from 66. 85% to 131. 74%. The relative standard deviations( RSD) ranged from 7. 39% to 9. 98%. In 30 seafood samples of 5 categories,small yellow croaker body had the highest unsaturated ω- 3 fatty acids level. Conclusion The method is simple,time- saving,with good linearity,it can be applied to detect the contents of 37 fatty acids in various seafoods,and provide technical support for monitoring and evaluation of nutrient contents in seafoods.
