白芍初加工过程中单萜苷类及多羟基化合物的变化分析 被引量:14

Analysis of Variation of Monoterpene Glycosides and Polyhydroxy Compounds in Paeoniae Radix Alba During Preliminary Processing

摘要 目的:探索不同加工干燥方法下,白芍中单萜苷类及多羟基化合物的变化规律。方法:新鲜白芍洗净,除去头尾及细根,晾干,分别经去皮切片、带皮切片、去皮水煮至透心切片、带皮水煮至透心切片、带皮水煮至透心后去皮切片、去皮隔水蒸至透心切片、带皮隔水蒸至透心切片、带皮隔水蒸至透心后去皮切片处理后,分别经35、45、60、80、100、120℃恒温鼓风烘干、晒干及阴干至含水量符合中国药典标准。HPLC-PDA法同时测定其中单萜苷类化合物(芍药苷、芍药内酯苷、氧化芍药苷、苯甲酰芍药苷)、多羟基化合物(儿茶素、没食子酸)及苯甲酸含量,进行主成分分析。结果:鲜白芍带皮处理比去皮处理综合得分高;鲜白芍经蒸制或水煮后切片干燥处理,单萜苷类及多羟基化合物含量增加,苯甲酸含量降低;鲜白芍经蒸制和水煮高温处理后,其后的干燥温度变化对其成分变化影响不大。结论:白芍初加工过程中,不同加工干燥方法对白芍活性成分有一定的影响。采用水煮处理后切片干燥再去皮的加工方法可得到较好品质的白芍加工品,但考虑到干燥后去皮的不易操作性,建议采用去皮切片后再干燥的方法。 Objective:To investigate variation of monoterpene glycosides and polyhydroxy compounds in Paeoniae Radix Alba dried by different processing methods. Methods:The crude drugs were processed sequentially as washed, removed the head, tail, fine roots and dried. The samples were divided into eight groups by whether peeled and decocted or not. Each group was dried by 35,45,60,80,100, 120 ℃ ,sun-dried and shade-dried. HPLC-PDA method was adopted to determine the content of monoterpene glycosides compounds (paeoniflorin alibiflorin, oxypaeoniflorin and benzoylpaeoniflorin ), polyhydroxy compounds (catechin and gallic acid ) and benzoic acid. Chromatographic conditions : Phecad C18 column ( 250 mm× 4. 6 mm,5 μm). A principal component analysis ( PCA ) method was used subsequently to get data processed. Results:The retained content of seven constituents decreased in those peeled crude drug, and after cooked,monoterpene glycosides and polyhydroxy compounds increased while the benzoic acid decreased. It was believed that rele- vant enzymes were inactivated while being cooked so that drying temperature showed little influence on the biotransformation. Conclusion : Contents of effective ingredients in Paeoniae Radix Alba are influenced by drying processing. The prefera- ble method shows to be that crude drug should be cooked before being peeled and dried. As a matter of processing convtence, it is sug- gested to be peeled and sliced before being dried.
