摘要
[目的]建立枳壳离体培养再生体系。[方法]采用均匀设计研究了不同时期枳壳茎尖、茎段、芽的愈伤组织诱导、增殖、分化、及生根情况。[结果]愈伤组织诱导率及生长量由高到低为春梢>夏梢>秋梢>冬梢;不同时期外植体诱导愈伤组织所需生长调节物质种类、浓度、配比不同,春梢茎段外植体诱导愈伤组织以NAA 0.2 mg/L+2,4-D 0.5 mg/L+KT 0.2 mg/L+IBA 0.1 mg/L为最好,夏梢茎段外植体诱导愈伤组织以NAA 0.2 mg/L+2,4-D 4.5 mg/L+KT 0.2 mg/L+IBA 0.1 mg/L为最好。愈伤组织增殖的最优配方是MT+6-BA 0.25 mg/L+NAA 0.2 mg/L。在MT+6-BA 0.25 mg/L+IBA 0.3 mg/L培养基上不定芽分化效果好,不定根发生率、根数、根的质量均在1/2MT+NAA 4 mg/L培养条件下最佳。[结论]为枳壳快速繁殖、脱毒技术和相关细胞工程的研究奠定基础。
[Objective] The aim was to study the establishment of regeneration system of Poncirus trifoliate(L.) Raf.[Method] A regeneration method via callus was established in which uniform design was employed in the experimentation.Induction rate and quantity of callus were influenced by the types of explants.[Result] Callus were induced satisfactorily with spring shoots cultured in MT medium supplemented with NAA 0.2 mg/L,2.4-D 0.5 mg/L,KT 0.2 mg/L and IBA 0.1 mg/L and summer shoots in MT mediuin supplemented with NAA 0.2 mg/L,2,4-D 4.5 mg/L,KT 0.2 mg/L and IBA 0.1 mg/L.The calluses underwent good proliferation in MT medium supplied 6-BA 0.25 mg/L and NAA 0.2 mg/L.MT medium with 6-BA 0.25 mg/L and IBA 0.3 mg/L was favourable for redifferentiation and 1/2MT medium with NAA 4 mg/L resulted in the formation of strong roots.[Conclusion] The study laid foundation for rapid seedling breeding,detoxification technology.
