摘要
目的:探讨微小RNA-29b(microRNA-29b,miR-29b)在舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)组织中的表达及其生物学意义。方法:收集2012-01-02-2013-03-15江西省肿瘤医院头颈外科行手术切除的45例TSCC及对应癌旁组织(距离切缘>2cm),qRT-PCR技术检测miR-29b在TSCC及对应癌旁组织中的表达情况,并分析其与临床病理特征的关系;应用miR-29bmimic转染人舌鳞癌Tca-8113细胞,MTT和流式细胞技术检测转染后细胞增殖和凋亡变化,qRT-PCR和蛋白质印迹法检测miR-29b潜在靶点Bcl-2mRNA及蛋白的表达变化。结果:qRT-PCR结果显示,miR-29b在TSCC组织中的表达量为0.213±0.071,显著低于对应癌旁组织的1.873±0.114,t=2.531,P=0.031;miR-29b低表达与肿瘤体积增大(≥3cm,χ2=4.022,P=0.033)及高TNM分期(Ⅲ+Ⅳ,χ2=3.398,P=0.036)具有显著相关性;MTT结果显示,在Tca-8113细胞中转染miR-29b48h后可显著降低细胞增殖能力,P<0.05;同时,转染miR-29b72h后细胞凋亡率由(2.31±0.98)%增加到(11.83±1.36)%,t=2.237,P=0.034。Real-time PCR及蛋白质印迹结果显示,转染miR-29b后可显著下调Tca-8113细胞内Bcl-2mRNA及蛋白表达,P<0.05。结论:TSCC组织中miR-29b表达下调并与其临床病理特征相关,miR-29b可能通过下调Bcl-2的表达来抑制舌鳞癌Tca-8113细胞增殖并促进其凋亡。
OBJECTIVE:To discuss the expression of microRNA-29b (miR-29b) and its role in tongue squamous cell carcinoma (TSCC). METHODS: We freshly collected 45 samples of surgically resected paired TSCC and normal tumor-ad- jacent tissues from 2012.1 to 2013.3 and detected miR-29b expression in the samples using qRT-PCR. The correlation be- tween miR-29b expression and clinicopathological features was analyzed. The miR-29b mimic was transfected into Tca- 8113 cells,cell proliferative ability and apoptosis were analyzed by MTT and Flow cytometry. Bcl-2 and potential targets of miR-29b were detected by qRT-PCR and Western blot. RESULTS: The expression of miR-29b in TSCC tissues was signif- icantly downregulated compared to those of the normal tumor-adjacent tissues (0. 213 ± 0. 071 vs 1. 873 ± 0. 114, t = 2. 531 ,P=0. 031). Down-regulation of miR-218 was found to correlate significantly with the larger tumor size (≥3 cm, x2 = 4. 022, P= 0. 033) and advanced TNM stage ( Ⅲ + IV, x2 = 3. 398, P = 0. 036). Upregulated the expression of miR- 29b in Tca-8113 cells could decrease the proliferation (P〈0.05), and enhance cell apoptosis (11.83±1.36) Mvs (2.31± 0.98) % ,t= 2. 237, P= 0. 034. Overexpression of miR-29b in Tca-8113 cells could downregulated both the mRNA and protein level of Bcl-2 (P〈0.05). CONCLUSION: The expression of miR-29b is downregulated in TSCC tissues and corre- lated with malignant clinicopathological characteristics of TSCC, and miR-29b may inhibit cell proliferation and promote cell apoptosis in TSCC by downregulating Bcl-2.
