摘要
从吉尔吉斯白桦转录组文库测序中获得白桦BkWRKY1转录因子的cDNA序列,该序列包含2 189个碱基。序列比对和同源性分析表明,该基因cDNA序列包含1 728 bp的开放阅读框,可编码575个氨基酸,5'非翻译区(UTR)为119 bp,3'非翻译区(UTR)为342bp。该基因属于WRKY transcription factor家族,含有2个WRKY结构域,蛋白分子量为62.483 kD,理论等电点为7.32,负电荷残基(Asp+Glu)总数为62个,正电荷残基(Arg+Lys)总数为62个。蛋白不含有信号肽,具有一定的亲水性,为亲水蛋白,无跨膜结构。同源性比较与进化树分析表明,吉尔吉斯白桦WRKY蛋白与大豆和蒺藜苜蓿的WRKY蛋白在进化上关系较近。该基因在0.6%NaHCO3胁迫处理后表达量增加,为上调表达基因。
The eDNA setltlemce of BkWRKYI transcription factor gene was obtained from Betula kirghisorum transcriptome sequencing libraryand the sequence consists of 2189 hp nucleotides, Sequence alignment and homology analysis showed that the gene eDNA sequence contains an open reading frame of 1728 bp, encoding 575 amino acids, 5 :untranslated region (UTR) 119 bp, 3 :untranslated region(UTR) 342 bp. The gene belongs tothe WRKY lranscription factor family, containing two WRKY domains. Prote.in molecular weight is 62. 483 kD and its the-oretical isoelectric point is 7.32. Negatively charged residues ( Asp + Glu) is a total of 62 and the total number of positively charged residues ( Arg + Lys) is 62. BkWRKY1 prolein has m) signal peptide and transmembrane domain, and has eertain hydrophilic, which is hydrophilic protein.homology comparison and phylogenetic analysis showed that Betula kirghisorum WRKY protein has closer ew+lutionary relationship with Glycine max and Medicago truncatula. BkWRKYI gene expression is increased after O. 6% NaHCO3 stress treatment, which is up-regula- led expression gene
