摘要
考察了脂肪酶对典型动物油脂(猪油、羊油、牛油)、植物油脂(橄榄油、大豆油、菜籽油)底物的酶解特性,以改进型铜皂-分光光度法测定的脂肪酶水解后体系中的脂肪酸含量表征酶活力大小。结果显示,碱性脂肪酶Leveking-P1000水解油脂的最适pH为8.0,不受底物来源的影响。脂肪酶对动物油脂的水解,受温度影响较大,当温度超过动物油脂的熔点时,脂肪酶对油脂的水解效率显著提高。油脂成分中不饱和脂肪酸越多,越有利于被脂肪酶水解。三种类型的表面活性剂SDS、1231和TX-10对脂肪酶水解不同油脂底物的影响结果相似,即混合乳化体系中表面活性剂浓度在0.2 mg/mL以下时,对脂肪酶水解猪油、牛油和菜籽油均表现出一定的激活作用,而浓度超过0.5 mg/mL则对脂肪酶水解油脂均表现为显著的抑制作用。
The characteristic of lipase hydrolyzing different triglycerides was investigated by using several typical greases and oils from animals and vegetables. The lipase activities measured by the modified colorimetric assay were applied to determine the amount of the fatty acid in the hydrolyzed emulsion. The results illustrated that the optimal application pH for Leveking-P1000 lipase is at pH 8.0 for all the origin greases and oils. The hydrolysis of animal greases by lipase was dramatically influenced by temperature. The efficiency for hydrolyzing animal greases could be improved when the temper- ature is above theirs melting temperature due to a better dispersion could be achieved. Three kind of surfactants SDS, 1231 and TX-10 showed the similar effect on hydrolysis of lipase with different origin greases and oils. In the lipase and substrate system, while the concentration of the surfactants is less than 0.2 mg/mL, the hydrolysis reaction could be acti- vated by the surfactants for hydrolyzing lard oil, beef tallow and colza oil; while the concentration of the surfactants is higher than 0.5 mg/mL, the hydrolysis reaction will be sharply inhibited for all the origin greases and oils.
