摘要
目的:建立西黄滴丸的高效液相色谱指纹图谱并进行指标成分胆红素的定量分析,为整体控制西黄滴丸质量和对天然牛黄定性定量检测提供依据。方法:色谱柱:Agilent ZORBAX SB-C18柱,4.6 mm×250 mm,粒度5μm,流动相为甲醇-0.1%磷酸溶液-乙腈(非线性梯度洗脱),指纹图谱检测波长239 nm;胆红素定量分析检测波长453 nm。结果:建立了10批西黄滴丸的对照指纹图谱,各批次样品的指纹图谱的相似度均大于0.98,确立共有峰11个,其中以胆红素峰为参照峰,并确定对胆红素进行定量分析的方法。结论:该方法操作简便,结果准确、可靠,可为西黄滴丸全面质量控制提供有效依据。
Objective :To establish high performance liquid chromatography(HPLC) fingerprint of Xihuang dripping pills and determine bilirubin of natural calculus bori to ensure the quality of Xihuang dripping pills. Methods:The fingerprint was obtained on an Agilent ZORBAX SB-C18column(4.6 mm×250 mm,5 μm)with the solvent system composed of methanol-0.1% phosphoric acidacetonitrile(nonlinear gradient elution).The detective wavelength was 239 nm and 453 nm. Results:In the established fingerprint,the simiarity between peaks was more than 0.98,total 11 co-possessing peaks were selected as the fingerprint peaks of Xihuang dripping pills when taking Bilirubin peak as the reference peak and make a quantitative analysis of the bilirubin. Conclusion:The method can be used for the quality control of Xihuang dripping pills with simple great precision and good reproducibility.
