hsa-mir-145-5p在尖锐湿疣组织中的表达及生物信息学分析 被引量:3

摘要 目的:检测hsa-mir-145-5p在尖锐湿疣(CA)组织中的表达,并分析该基因的生物信息学特征。方法:选取CA患者30例以及对照组20例,取材后提取组织总RNA,进行逆转录,荧光定量PCR检测hsa-mir-145-5p在各个标本中的表达。通过targetscan数据库分析hsa-mir-145-5p的靶基因,并通过DAVID数据库进行生物信息学分析。结果:hsa-mir-145-5p在尖锐湿疣组织中的相对表达量为0.20±0.25,相对对照组(1.76±0.61)降低约88.6%。生物信息学分析发现,多达58.9%的hsa-mir-145-5p的靶基因属于磷酸化蛋白,并有15.4%的靶基因参与转录调控,17.7%的靶基因参与乙酰化。此外,hsa-mir-145-5p的靶基因还属于MAPK信号通路、Wnt信号通路、TGF-beta信号通路等信号通路的信号分子。结论:hsa-mir-145-5p在尖锐湿疣组织中呈低表达特性。通过生物信息学分析,推测HPV可能通过诱导hsa-mir-145-5p的低表达,释放hsa-mir-145-5p对MAKP及其它信号通路相关蛋白翻译的抑制,从而激活相关信号通路。
