摘要
目的研究补阳还五汤(黄芪、当归、白芍、地龙、川芎、红花、桃仁)对失神经肌萎缩大鼠Angptl4基因表达的影响,探讨其对失神经肌萎缩的保护机制。方法建立大鼠腓总神经夹伤模型,将造模后的60只SPF级SD大鼠随机分为:假手术组、补阳还五汤高、中、低剂量组、弥可保组与模型组,术后每日灌胃给药。18 d后qRT-PCR与Western-blot检测Angptl4基因与蛋白的差异表达。结果与模型组相比,补阳还五汤中、高剂量组Angptl4 mRNA与蛋白表达均显著增高(P<0.05或P<0.01)。结论补阳还五汤可能通过增加Angptl4基因与蛋白表达,促进血管新生,改善失神经萎缩肌肉的血液供应,从而延缓肌萎缩的发生。
AIM To study the effect of Buyang Huanwu Decoction (Astragali Radix, Angelicae sinensis Ra- dix, Paeoniae Radix alba, Pheretima, Chuanxiong Rhizoma, Carthami Flos, Persicae Semen) on Angptl4 and pro- tein gene expression in the denervated tibial muscle atrophy of rats and their protection mechanism. METHODS After 60 Sprague-Dawley rats subjected to common peroneal nerve crush model of 5 mm injury, the rats were ran- domly divided into six groups for daily intragastric administration of drugs : sham operation group, the model group, high, medium, low dose of Buyang Huanwu Decoction groups, mecobalamin group (as a positive control). The drug administration lasted for 18 days. Different expression levels of Angptl4 in the tibial muscle of rats after the op- eration were performed by qRT-PCR and Western-blot. RESULTS Compared with the model group, Angptl4 mR- NA and protein expression in Buyang Huanwu Decoction medium and high-dose group increased significantly (P 〈 0. 05 or P 〈0.01 ). CONCLUSION Buyang Huanwu Decoction maybe increase Angptl4 expression to promote the angiogenesis and improve blood supply for the denervated tibial muscle atrophy of rats.
