摘要
目的研究肌成束蛋白Fascin-1在颞叶癫痫模型大鼠颞叶皮质及海马中的表达变化,探讨Fascin-1与颞叶癫痫的关系及其意义。方法 56只雄性SD大鼠分为对照组(5只)、实验组(35只)以及用于行为学观察的干预组(8只)和模型组(8只)。实验组大鼠腹腔注射匹鲁卡品建立颞叶癫痫模型,并在模型点燃后的不同时间点(6 h、24 h、72 h、1周、2周、1个月、2个月)断头取脑;干预组进行外源性Fascin-1干预后,观察其造模后行为学的改变。最后分别运用免疫组化、Western blot检测实验组及对照组颞叶皮质和海马中Fascin-1蛋白的动态变化;用免疫荧光双标对Fascin-1进行定位。结果行为学结果示干预组癫痫发作的潜伏期及达到4/5级所需的时间较模型组明显延长(P<0.05),但发作程度与模型组比较差异无统计学意义(P>0.05)。免疫组化结果显示,实验组大鼠颞叶皮质和海马中的Fascin-1表达水平在各个时间点都低于对照组,差异有统计学意义(P<0.05,皮质6 h时间点除外)。Western blot检测结果同样显示实验组Fascin-1蛋白表达水平在各个时间点都低于对照组,差异有统计学意义(P<0.05,皮质6 h时间点除外)。免疫荧光双标检测结果显示,Fascin-1主要表达于神经元上而不是星形胶质细胞。结论 Fascin-1蛋白的水平在颞叶癫痫大鼠模型中显著下调,外源性给予Fascin-1多肽后,发现对癫痫的发作有延缓作用。提示Fascin-1蛋白的降低可能与颞叶癫痫的病理过程有着密切关系。
Objective To determine the expression pattern of fascin-1 in the temporal lobe cortex and hippocampus of rat epilepsy model, and to investigate the role of its expression in temporal lobe epilepsy (TLE). Methods A total of 56 male SD rats were randomly divided into control group (n = 5 ), experimental group ( n = 35 ), intervention group ( exogenous fasein-1 of 0.05 μg/kg was given by intra- peritoneal injection for 5 consecutive days, n = 8) and epilepsy model group ( n = 8 ). The rats of 4 later groups were injected intra-peritoneally with pilocarpine to establish TLE model. The rats of intervention group were observed for their behavior changes and evaluated by Racine' s scoring system. The rats of control and experimental group were sacrificed at different time points, including 6, 24 and 72 h, 1 and 2 weeks, and 1 and 2 months after kindling. Immunochemical assay, Western blotting and immunofluorescenee assay were used to assess the expression and location of fascin-1 in the cortex and hippoeampus. Results The latent period and the time required to reach 4/5 level was prolonger in the intervention group than model group ( P 〈 0.05 ), but there was no significance in attack degree between the 2 groups. In the hippocampus and adjacent cortex in epileptic rats, fascin-1 expression was significantly decreased at all time points after kindling when compared with the controls (P 〈 0.05, but not at the time point of 6 h in the cortex). Immnofluorescence assay showed fascin-1 was mainly expressed in the neurons but not in the astrocytes. Conclusion The expression pattern of fascin-1 is down-regulated in the TLE rat model, and epilepsy seizures are delayed after giving exogenous fascin-1 peptides, indicating that decreased expression of fascin-1 might be involved in the pathogenesis of TLE.
