柴胡疏肝散对癫痫合并抑郁大鼠的行为学及海马齿状回神经发生的影响 被引量:8

Effects of Chaihu-Shugan-San on Behavior and Neurogenesis of Hippocampal Dentate Gyrus in Epileptic Rats with Depression

摘要 目的:探讨柴胡疏肝散(CHSGS)对慢性癫痫合并抑郁大鼠的行为学及海马齿状回神经发生的影响。方法:24只癫痫合并抑郁(锂-匹罗卡品诱导)大鼠随机分为模型组、氟西汀组和CHSGS组,每组8只,另设置正常对照组(n=8)。造模第21天后正常对照组予生理盐水10 mL/kg,氟西汀组每日灌服氟西汀(0.75mg/kg);CHSGS组每日灌服2.7 g生药/kg,连续1月。利用糖水消耗试验、旷场试验及免疫组织化学方法检测大鼠的行为学及海马齿状回神经发生改变。结果:与正常对照组比较,模型组、氟西汀组、CHSGS组的糖水消耗及行为学评分明显下降,海马齿状回新生神经元增加(P<0.05)。氟西汀组、CHSGS组糖水消耗及行为学评分较模型组明显提高,海马齿状回新生神经元增加(P<0.05);氟西汀组、CHSGS组相比差异无统计学意义。结论:CHSGS能提高癫痫合并抑郁大鼠海马齿状回的神经发生,改善抑郁症状。 Objective: To explore the effects of Chaihu-Shugan-San (CHSGS) on behavior and neurogenesis of the hippocampal dentate gyrus in chronic epileptic rats with depression. Methods: Twenty-four rats with depression which were selected from the chronic pilocarpine-induced epileptic rats were randomly divided into 3 groups (8 in each): the groups of model, Fluoxetine and CHSGS treatment. And the normal control group was established (n=8). After 21 d ofpilocarpine-induced epilepsy in rats, the model group received saline (10 mL/kg), the Fluoxetine group was given Fluoxetine (0.75 mg/kg), and the CHSGS group was given CHSGS (2.7 g/kg) for 1 month. Saccharose water consumption test, open-field test and immunohistochemistry were used to observe the behavior and neurogenesis in the hippocampal dentate gyrus. Results: when compared with those in the normal control group, the consumption of saccharose water and behavioral scores were significantly decreased whereas the number of newborn neurons in the hippocampal dentate gyrus were increased in the other three groups (P〈 0.05). The consumption of saccharose water, behavioral scores and numbers of newborn neurons in the hippocampal dentate gyrus were increased remarkably in the fluoxetine group and CHSGS group than than those in the model group (P〈0.05). There were no significant differences between the fluoxetine group and CHSGS group. Conclusion: CHSGS can increase the neurogenesis of the hippocampal dentate gyrus in epileptic rats with depression and effectively improve behavior recovery.
