银屑病患者皮肤间充质干细胞中表皮生长因子、转化生长因子-β1的表达水平及其意义 被引量:3

Expression levels and significance of EGF,TGF-β1 of skin mesenchymal stem cells in patients with psoriasis

摘要 目的观察银屑病患者皮肤间充质干细胞(SMSCs)的形态特征、免疫表型、诱导分化情况及其分泌表皮生长因子(EGF)及转化生长因子-β1(TGF-β1)的水平,尝试探讨银屑病可能的发病机制。方法选取35例银屑病及35例非银屑病患者设为观察组及对照组,分离、培养2组人群的SMSCs,流式细胞仪鉴定免疫表型,多向分化法鉴定细胞分化功能,ELISA法测定EGF、TGF-β1的表达水平。结果 2组患者的SMSCs形态学特征、免疫表型及诱导分化结果比较相似,观察组SMSCs分泌EGF含量显著高于对照组(P<0.01),TGF-β1含量显著低于对照组(P<0.01),且2种细胞因子的含量高低与患者病情严重程度无相关性(P>0.05)。结论银屑病患者的SMSCs分泌EGF、TGF-β1的水平异常,可能参与了银屑病的发病机制。 Objective To observe morphological characteristics,immunophenotyping,differentiation and the expression level of epidermal growth factor(EGF) and transforming growth factor-β1(TGF-β1) in skin mesenchymal stem cells(SMSCs) so as to explore the possible mechanism of psoriasis. Methods 35 patients with psoriasis and 35 patients with non-psoriasis were selected and designed as observation group and control group. SMSCs of 2 groups were separated and cultured. Immunophenotyping was determined by flow cytometry(FCM). Cell differentiation ability was evaluated by multidirectional differentiation method. The expression level of EGF and TGF-β1 was detected by ELISA. Results Morphological characteristics,immunophenotyping,differentiation of SMSCs were similar. The expression level of EGF in observation group was higher than control group(P 0. 01),but the expression level of TGF-β1 was significantly lower than control group(P 0. 01). In addition,the expression level of EGF and TGF-β1 in 2 groups had no significant relationship with the severity of disease(P 0. 05). Conclusion The expression level of EGF and TGF-β1 is abnormal in patients with psoriasis,which may participates in the pathogenesis of psoriasis.
