摘要
研究从几种天然产物中筛选出有效的黄嘌呤氧化酶抑制剂。采用酶促反应动力学的方法研究绞股蓝皂苷、葛根素、苹果多酚、褐藻糖胶4种天然产物的黄嘌呤氧化酶体外抑制作用,进行动态监测,绘制LineweaverBurk曲线,计算IC50值,探讨其抑制效果和作用方式。试验结果表明,苹果多酚可以通过反竞争性抑制作用,有效抑制黄嘌呤氧化酶活性;褐藻糖胶虽在体外模型中也表现出显著抑制黄嘌呤氧化酶活性的作用,但抑制类型不典型。上述两种活性物质是来源于普通食品中的天然产物,具有安全可靠的特性,可以作为改善高尿酸血症患者健康状况的保健类食品原料。
Xanthine oxidase is the key enzyme in uric acid generation process. And uric acid induce the hyperuricemia which is the increased risk of metabolic syndrome in recent years. This study aimed to screening of xanthine oxidase inhibitors from several natural products such as gypenoside, puerarin, apple polyphenols and fucoidan. The traditional method in enzymatic reaction kinetics including dynamic monitoring, Lineweaver-Burk curve drawing and calculation of IC50 were used to research the inhibition effect and effect way in the investigation. The results showed that apple polyphenol could effectively inhibit the activity of xanthine oxidase via anti-competitive mode, while fucoidan also showed significant inhibition of the activity of xanthine oxidase in vitro model without typical mode. The two kinds of active substances above are natural product derived from the ordinary food, with safe and reliable characteristics, can be used as materials of health food to improve the health status of patients with hyperuricemia or gout.
