摘要
以耐铜性较强的烟草W38和耐铜性较弱的本氏烟为对比试验材料,通过水培法探究4mg/L铜胁迫下不同浓度(0,100,300,500μmol/L)水杨酸(SA)对2个烟草品种碳代谢的相关物质含量及几个关键酶的影响。结果表明:经过4mg/L铜处理15d后,烟草W38和本氏烟均受到一定的抑制,叶绿素a、b含量、葡萄糖、果糖、蔗糖含量下降,蔗糖磷酸合成酶(SPS)、蔗糖合成酶(SS)和淀粉酶的活性显著降低(p<0.05),而淀粉含量却呈现出相反的变化趋势,显示出2个烟草品种耐铜性差异,淀粉酶、酸性转化酶(AI)、中性转化酶(NI)活性均升高。而喷施适宜浓度的SA能够显著提高植株叶绿素a、b含量、葡萄糖、果糖、蔗糖含量和SPS、SS、淀粉酶活性,而烟草淀粉含量和AI、NI活性却呈现出下降趋势。由此可知,适宜浓度的SA有缓解烟草铜胁迫的效果,但是不能消除铜胁迫对烟草生长的抑制作用,外源SA缓解烟草铜胁迫的最适浓度为300μmol/L,且高浓度的SA不但不能缓解铜胁迫对烟草的抑制作用,而且会对烟草有一定的毒害作用。
A solution culture experiment was conducted to study the effects of different salicylic acid (SA) concentration (0,100,300,500μmol/L) on carbon metabolism of two tobacco cultivars, Nicotiana W38 (Cu resistance genotype) and Nicotiana benthamiana (Cu sensitive genotype) under copper stress. The results showed that: The tobacco seedlings under 4 mg/L Cu Stress for 15 days, the Chl a, Chl b, glucose, fruc- tose, sucrose content and SPS,SS, amylase activity significantly decreased(p〈0.05) of the two tobacco cul- tivars. The starch content ratio changed in the opposite trend, indicating the differences of the two tobacco cultivars in Cu tolerance. The amylase, AI, NI activity increased. Spraying suitable concentration of SA could improve the content of Chl a, Chl b, glucose, fructose, sucrose and the activity of amylase, AI, NI and reduce the starch content and the activity of AI, NI. 300μmol/L was the optimal concentration of exoge- nous SA to relieve Cu stress of the two tobacco cultivars. SA could alleviate the effect of Cu stress, but it could not eliminate the inhibitory effect of Cu stress on the growth of tobacco, while high concentration of SA might even produce a new stress affection of tobacen
