鼻咽癌组织中ERK1/2、p-ERK1/2和MMP-9蛋白的表达变化及其相关性 被引量:5

摘要 目的观察细胞外调节蛋白激酶1/2(ERK1/2)、磷酸化细胞外调节蛋白激酶1/2(p-ERK1/2)和基质金属蛋白酶9(MMP-9)在鼻咽癌(NPC)组织中的表达及相关性,并探讨其意义。方法采用免疫组织化学染色SP法检测75份NPC组织(NPC组)和27份鼻咽黏膜慢性炎组织(炎性组)中ERK1/2、p-ERK1/2和MMP-9蛋白的表达,分析三者在NPC组织中表达的相关性。结果 NPC组ERK1/2、p-ERK1/2和MMP-9蛋白阳性表达率均显著高于炎性组(P均<0.01),χ2值依次为19.8、17.1、21.3;NPC组织中ERK1/2、p-ERK1/2和MMP-9蛋白三者间的表达呈正相关(P均<0.01),其中ERK1/2与p-ERK1/2、MMP-9蛋白的r值分别为0.58、0.34,p-ERK1/2、MMP-9蛋白的r值为0.34。结论 ERK1/2、p-ERK1/2和MMP-9蛋白在NPC组织中呈高表达且三者间呈正相关,p-ERK1/2可能通过调节MMP-9表达参与NPC侵袭、转移。 Objective To investigate the expression changes of extracellular signal-regulated kinase 1/2 (ERK1/2), phosphorylated extracellular signal-regulated kinase 1/2 ( p-ERK1/2 ) and matrix metalloproteinase 9 ( MMP-9 ) in naso-pharyngeal carcinoma (NPC) and their correlations, and to discussed the significance.Methods The expression of ERK1/2, p-ERK1/2 and MMP-9 proteins was detected in 75 cases of NPC tissues ( NPC group) and 27 cases of chronic nasopharyngeal inflammation tissues (CNI group) by immunohistochemical SP method , and the relationships among them in NPC tissues were analyzed .Results The positive expression rates of ERK 1/2, p-ER-K1/2 and MMP-9 proteins of the NPC group were significantly higher than those of CNI group (all P〈0.01, χ2 =19.8, 17.1 and 21.3, respectively). The expression of p-ERK1/2 and MMP-9 was positively correlated with ERK1/2 (r=0.58,0.34 respectively, both P〈0.01), p-ERK1/2 and MMP-9 expression was positively correlated with each other (r=0.34, P〈0.01).Conclusion ERK1/2, p-ERK and MMP-9 proteins are highly expressed in human NPC and is positively correlated with each other .The p-ERK1/2 may participate in the invasion and metastasis of NPC by regulating the expression of MMP-9.
