枸杞多糖对低剂量辐照雄性大鼠性功能损伤恢复的影响 被引量:9

EFFECT OF LYCIUM BARBARUM POLYSACCARIDES ON RECOVERY OF DAMAGED SEXUAL FUNCTION INDUCED BY LOW DOSAGE OF IONIZING RADIATION IN MALE RATS

摘要 目的探讨枸杞多糖(Lycuim barbarum polysaccharide,LBP)对低剂量辐照雄性大鼠性功能损伤自然恢复的影响。方法将Wistar雄性大鼠随机分为:正常对照、辐射对照1、辐射对照2、辐射对照3、LBP+辐射1、LBP+辐射2、LBP+辐射3共7组,每组12只。除正常对照组外,其余各组于周1至周5用60Coγ射线治疗机对大鼠睾丸组织进行局部辐射,连续4w。辐射剂量:0.015Gy/(capita·d),总剂量:2.3Gy/capita。每次辐射前,LBP+辐射1、2、3组给予10mg/kg LBP进行灌胃干预,辐射对照1、2、3灌胃等体积的生理盐水。分别于末次辐射后1、7和14d处死,观察雄性大鼠阴茎勃起功能和性功能的变化,计数精子数、活率,测定睾丸、附睾、前列腺和精囊腺脏器系数及血清中睾酮(testosterone,T)、促黄体生成激素(luteinizing hormone,LH)和促卵泡生成激素(follicle stimulating hormone,FSH)的水平。结果与辐射对照1、2、3组相比,LBP+辐射1、2、3组的大鼠阴茎勃起潜伏期、扑捉潜伏期和射精潜伏期明显缩短,扑捉次数和射精次数明显增加;睾丸脏器系数、精子计数及活率明显提高;血清中性激素T水平显著上升。结论 LBP可以明显改善低剂量辐照损伤后雄性大鼠的性功能,对雄性大鼠辐照损伤后性功能的恢复具有促进作用。 Objectives To investigate the effect of Lycium barbarum polysaccbarides (LBP) on damaged sexual function in male rats inflicted with low dosage of ionizing radiation. Methods Wistar rats were randomly divided into seven groups: normal control group; radiation control groupl, 2 and 3; LBP+radiation group 1, 2, and 3(n=12 for each group). Except the normal control group, the other groups were exposed to local radiation using a 60Coγ irradiation therapy machine from Monday to Friday each week for four consecutive weeks (total dose of 2.3Gy for each rat). For LBP+radiation group 1, 2 and 3, gastric gavage with LBP (10mg/kg) was given prior to the radiation exposure. For radiation control groupl, 2 and 3, the rats were given gastric gavage with same volume of saline. The rats were sacrificed on days 1, 7 and 14 after the last exposure. The erectile function and sexual function were observed. Finally, the organ coefficients of testis, epididymis, seminal vesicle-prostate and the sperm count and viability were determined. The levels of sex hormone T, LH and FSH in serum were measured. Results Compared with radiation control group 1,2 and 3, the erection latency, mounting latency and ejaculation latency of the rats treated with LBP were reduced significantly; the number of mountings and ejaculations increased significantly; testicular organ coefficients, sperm count, sperm viability promoted significantly, Meanwhile, the level of T in serum was increased. Conclusion LBP is able to improve the sexual function in male rats exposed to low dosage of ionizing radiation, and plays a vital role in promoting spontaneous recovery of sexual function in irradiated rats.
