新疆细虫草菌核的膨大弯颈霉菌株SFYT002核苷类成分分析 被引量:3

Determination of nucleosides of Tolypocladium inflatum strain SFYT002 isolated from sclerotium of Ophiocordyceps gracilis in Xinjiang

摘要 目的:定量分析膨大弯颈霉菌株SFYT002菌丝体中核苷类成分。方法:本研究从新疆细虫草菌核分离出一株膨大弯颈霉菌株SFYT002,以已知的7种核苷为标准品,并与野生新疆细虫草和冬虫夏草做比较,采用高效液相色谱法(HPLC)对其核苷类成分进行测定。结果:膨大弯颈霉SFYT002菌株含有与冬虫夏草相同的7种核苷类成分,其中该菌株尿嘧啶为75.668μg·g-1,略高于冬虫夏草,为新疆细虫草的4倍左右;肌苷含量达到663.776μg·g-1,为冬虫夏草3倍,为新疆细虫草的27倍之多;腺苷含量114.653μg·g-1,分别为冬虫夏草和新疆细虫草腺苷含量的1/5和2/5左右;尿苷、鸟苷、腺嘌呤含量也接近冬虫夏草,高于新疆细虫草。此外,在膨大弯颈霉SFYT002菌株中检测出虫草素,含量为32.261μg·g-1,但新疆细虫草和冬虫夏草中未检测出虫草素,为虫草素开发提供了一种新的药源。结论:膨大弯颈霉菌株SFYT002核苷类成分含量丰富,具有开发潜力和前景。 Objective: The nucleoside contents were quantitatively analyzed in the mycelium of Tolypocladium inflatum strain SFYT002. Methods: In the present study,a strain SFYT002 of Tolypocladium inflatum was isolated from the sclerotium of Ophiocordyceps gracilis in Xinjiang. Seven known chemical compounds of nucleosides were used as standard reference to determine the nucleoside contents of this strain using HPLC method,which were then compared with those of wild Ophiocordyceps gracilis and Ophiocordyceps sinensis. Results: The strain SFYT002 contained all of the seven nucleosides,which were previously found in Ophiocordyceps sinensis. Uracil was assayed to be75. 668 μg·g- 1in the strain SFYT002,which was higher than that in Ophiocordyceps sinensis and was 3 times higher than that in Ophiocordyceps gracilis. Inosine was tested to be 663. 776 μg·g- 1in the strain SFYT002,which was respectively 3 and 27 times higher than those in Ophiocordyceps sinensis and Ophiocordyceps gracilis. Adenosine was determined to be 114. 653 μg·g- 1in the strain SFYT002,which was respectively 1 /5 and 2 /5 of those in Ophiocordyceps sinensis and Ophiocordyceps gracilis. The contents of other nucleosides,such as uridine,guanosine and adenine,were similar to those in Ophiocordyceps sinensis,but higher than those in Ophiocordyceps gracilis. Furthermore,cordycepin was detected in the strain SFYT002,with a content of 32. 261 μg·g- 1. Cordycepin was not detected in the samples of Ophiocordyceps sinensis and Ophiocordyceps gracilis. The strain SFYT002 would provide a new resource of cordycepin for drug development. Conclusion: The strain SFYT002 of Tolypocladium inflatum has a good potential for developing nucleoside health food and medicine.
