摘要
目的探讨血必净对脓毒症大鼠炎症因子及肾组织中补体C3的影响。方法 90只SD大鼠随机均分为假手术组(SH组)、脓毒症组(SEP组)和血必净处理组(XBJ组),每组再分为2、6、8、12h和24h五个亚组,每亚组6只。采用盲肠结扎穿刺法建立脓毒症大鼠模型。术后SH组与SEP组经尾静脉给予生理盐水4ml/kg,每12小时1次;XBJ组给予血必净注射液4ml/kg,每12小时1次。术后各时间点采用双抗体夹心ELISA法检测血清中TNF-α、IL-6和IL-10水平,免疫组化及Western blot法检测肾组织中补体C3的表达。结果与SH组相比,SEP组与XBJ组各时间点血清TNF-α及IL-6浓度均升高(P<0.05),TNF-α出现双峰现象;XBJ组TNF-α及IL-6浓度低于SEP组(P<0.05),IL-10浓度高于SEP组(P<0.05)。与SH组相比,补体C3在SEP组及XBJ组中均有表达,且SEP组表达高于XBJ组(P<0.05)。结论血必净能降低血清TNF-α及IL-6水平,促进IL-10分泌,减少补体C3的表达,对脓毒症大鼠起保护作用。
Objective To investigate the effect of Xuebijing injection on inflammatory factors and complement C3 in kidney tissues of septic rats. Methods Ninety SD rats were equally randomized into three groups of SH （sham operated）, SEP and XBJ. Every group was then subdivided into five groups of 2,6,8,12 h and 24 h with 6 rats each. The septic rat model was made by cecal ligation and puncture in groups of SEP and XBJ. After operation, the rats in groups of SH and SEP were given normal saline 4 ml/kg every 12 hours,and those in group XBJ were given Xuebijing injection 4 ml/kg every 12 hours via the tail vein. Serum levels of TNF-α, IL-6 and IL-10 were detected by double antibody sandwich ELISA and the expression of complement C3 in kidney tissues was detected by immunohistochemistry and Western blot at the time points mentioned above after operation. Results Compared with group SH, serum concentrations of TNF-α and IL-6 in groups of SEP and XBJ were significantly increased at each time point（P〈0. 05）, which of TNF-a showed a bimodal phenomenon. Serum concentrations of TNF-α and IL-6 were significantly lower, while IL-10 was significantly higher,in group XBJ than those in group SEP（P〈0. 05）. Compared with group SH, complement C3 was expressed in groups of SEP and XBJ, which in group SEP was significantly higher than that in group XBJ （P〈0. 05）. Conclusion Xuebijing injection can protect septic rats by reducing serum levels of TNF-α and IL-6, promoting the secretion of IL-10, and decreasing the expression of complement C3.
