摘要
采用高效液相色谱串联质谱法测定鲫鱼血浆中喹乙醇的浓度。选用健康鲫鱼,随机分成2组,每组20尾,以30mg/kg体重的剂量经口灌服给药,血浆用30g/L的三氯乙酸沉淀蛋白后测定其浓度,统计软件3P97处理血药浓度-时间数据。鲫鱼经口灌服喹乙醇后的数据符合一级吸收一室模型,主要药动学参数:T1/2kα为1.09h±0.14h;T1/2Ke为5.97h±1.06h;Tmax为3.25h±0.41h;Cmax为25.73mg/L±2.53mg/L;AUC为325.35mg/(L·h)-1±69.01。鲫鱼口服喹乙醇吸收迅速完全,消除较慢。
The concentration of olaquindox in the plasma of Carassius auratus was determined by high performance liquid chromatography tandem mass spectrometry (LC-MS/MS) . Healthy Carassius auratus, were randomly divided into 2 groups,20 tail each group of , 30 mg per kg body weight oral dose of olaquin- dox was used,the blood samples were collected. The plasma was treated with 30 mL/L trichloroacetic acid concentrations for precipitation and detection of protein, statistical software 3P97 was used for processing plasma concentration-time data. It' s best to fit the olaquindox concentration-time data to one-compartment open model with first order absorption after single oral dosing. The pharmacokinetic parameters of ola- quindox in Carassius auratus are as follows : the elimination half-life T1/2Ke is 5.97 h± 1.06 h , the distri- bution half-life T1/2k. is 1.09 h±0.14 h , the peak time Tmax is 3. 25 h±0.41 h , the peak concentration CTmax is 25.73 mg/L±2.53 mg/L,and the area under the curve (AUC) is 325.35 mg/(L · h)^-1 ±69.01 mg/(L ·h)^-1 The results showed that olaquindox was rapidly and completely absorbed and slowly elimi- nated after oral administration to Carassius auratus.
