摘要
目的:体外观察人参皂苷联合地塞米松(DEX)对原代淋巴细胞白血病细胞凋亡和P-糖蛋白表达的影响。方法:抽取临床复发耐药的急性淋巴细胞白血病患者骨髓单个核细胞,体外培养,随机设对照组、人参皂苷组、DEX组、人参皂苷加DEX组。通过锥虫蓝拒染法测定细胞生长抑制率,流式细胞术测定细胞凋亡率及P-糖蛋白的表达。结果:细胞培养72 h后,人参皂苷组、DEX组、人参皂苷加DEX组对细胞生长均有抑制作用,且可导致细胞凋亡的发生,以人参皂苷加DEX组最为显著,并可以显著降低P-糖蛋白的表达。结论:人参皂苷联合DEX具有增强DEX诱导原代淋巴细胞白血病细胞凋亡的作用。
Objective : To explore the influence of Ginsenosides plus Dexamethasone on apoptosis and P-gp expression of primary lymphocytic leukemia cell. Methods:Primary leukemic cells were incubated with Ginsenosides, Dexamethasone and Ginsenosides plus Dexamethasone. The inhibition rate was observed by Trypan blue staining. The cell apoptosis rates and P-gp expression were detected by cytometer with Annexin-V/PI. Results:The results showed that either Ginsenosides therapy or Dexamethasone could inhibit the proliferation of leukemia ceils. Ginsenosides combined with Dexamethasone had stronger effect on primary leukemic cells. And Ginsenosides combined with Dexamethason also was stronger than that of the single Ginsenosides or Dexamethasone in inducting apoptosis and downregulating P-gp expression( P 〈 0.05 ). Conclusion: Ginsenosides plus Dexamethasone could reinforce the effect of Dexamethasone for cell apoptosis.
