摘要
目的:研究芹菜素(Apigenin,APG)对大鼠永久性局灶性脑缺血的保护作用。方法:采用线栓法建立大鼠永久性大脑中动脉阻塞(permanent middle cerebral artery occlusion,pMCAO)模型。将动物随机分为假手术组、缺血对照组、芹菜素(10、25和50mg/kg)治疗组、金纳多4mg/kg治疗组,插入栓线后立即腹腔注射给药。术后6h,采用盲法进行神经功能评分,测定脑梗死体积(IV%)、脑含水量及观察脑组织形态学改变,采用荧光标记法测定神经细胞胞浆Ca2+浓度([Ca2+]i)。结果:芹菜素组脑缺血大鼠行为学体征显著改善、脑梗死体积显著减少、脑组织含水量显著降低,脑组织形态学改变显著减轻,胞浆[Ca2+]i浓度显著降低。其中,芹菜素25mg/kg组治疗效果最为显著。结论:芹菜素对大鼠永久性局灶性脑缺血损伤具有保护作用。
Objective: To investigate the protective effects of apigenin( APG) against permanent focal cerebral ischemic injury in rats. Methods :The model of permanent middle cerebral artery occlusion( pMCAO) was established with Zea-permanent middle cerebral artery occlusion( pMCAO) was established with Zea-Longa occluding suture in rats. Which were randomly devided into six groups: sham operation group,ischemia control group,APG 10,25 and 50 mg /kg treated groups,and Ginaton 4 mg /kg treated group. Drugs were given immediately by intraperitoneal injection when the suture was inserted. At 6h after middle cerebral artery occlusion surgery,the neurological deficits,ratio of infarct volume,water content of the brain and histopathological changes were evaluated. The intracellular calcium concentration in neurons was determined by fluorescence labeling. Results: The neurological scores / ratio of infarct volume / brain water content / the intracellular calcium concentration of APG-treated groups were significantly decreased,and the histopathological changes were significantly improved compared with ischemia control group. Especially,the treatment effect of APG 25mg /kg groups was the most significant. Conclusion: APG had protective effects against permanent focal cerebral ischemic injury in rats.
