摘要
目的:探讨白藜芦醇(RES)对膀胱移行细胞癌T24细胞凋亡及组织蛋白酶B(CB)表达的影响。方法:取对数生长期的T24细胞,RES(25μmol/L)处理后分别培养24、48、72 h,以不作处理的细胞作空白对照。应用流式细胞术检测T24细胞的凋亡情况,RT-PCR法检测CB mRNA的表达情况,Western blot方法检测CB蛋白的表达情况。结果:RES作用后,实验组T24细胞凋亡增加,CB mRNA和蛋白的表达增强,均呈时间依赖性(F组间=20.742、27.767、38.750,F时间=164.963、22.399、27.912,F交互=63.745、76.532、91.722,P均<0.001)。结论:CB可能参与了RES诱导膀胱移行细胞癌T24细胞的凋亡。
Aim: To investigate effect of resveratrol( RES) on cell apoptosis and expression of cathepsin B( CB) in T24 cells. Methods: T24 cells induced by RES in logarithmic growth phase was selected and after cultured for 24,48 and72 h,respectively. The apoptosis of T24 cells was detected by flow cytometry. The expression of Caspase-9 mRNA was examined by RT-PCR and CB protein by western blot. The blank control group was the media without RES. Results: Apoptosis of T24 cells acted by RES apparently increased with time. It depended on time and was statistically significant. Expression of Caspase-9 mRNA apparently increased with RES acting time. It depended on time and was statistically significant. Expression of CB protein apparently increased with RES acting time. It depended on time and was statistically significant( Fgroup= 20. 742,27. 767,38. 750,Ftime= 164. 963,22. 399,27. 912,Finteraction= 63. 745,76. 532,91. 722,P〈 0. 001).Conclusion: CB might play a role in the apoptosis of bladder transitional cell carcinoma T24 cells induced by RES.
