摘要
目的观察甘草煎剂对大鼠胃电活动、胃窦组织中5-羟色胺摄取和脱羧(5-HT APUD)细胞的数目、面积及平均灰度值的影响。方法将70只大鼠随机分为随机分为7组,每组10只,雌雄各半,适应性喂养3d后,模型组分别给予不同剂量(5g/kg、10g/kg、15g/kg、20g/kg、40g/kg)的甘草煎剂灌胃,连续给药5d。正常组给予等体积生理盐水灌胃。各组动物于末次给药前24h禁食不禁水。观察各组大鼠的胃电活动变化,并在处死大鼠后采用免疫组化法观测各组大鼠胃窦组织中5-HT APUD细胞的数目、面积及平均灰度值。结果小剂量的甘草煎剂能明显抑制大鼠的胃电活动,减少大鼠胃窦组织中的5-HT APUD细胞数目及面积,增加平均灰度值,与正常组比较具有显著差异性(P<0.01);随着剂量的加大,大鼠的胃电活动得到一定的改善,胃窦组织中5-HT APUD细胞数目及面积有所增加,5-HT APUD细胞的平均灰度值变小,但与正常组比较也有差异性(P<0.05)。结论一定剂量的甘草煎剂能建立胃动力低下大鼠模型,其机制可能是通过减少胃窦组织中5-HT APUD细胞数目和和面积,增加5-HT APUD的平均灰度值而实现的。
Objective To observe the effect of different doses of the liquorice decoction on the gastric electrical activity and 5 - HT APUD cell number,dimension,average grey values in low gastric motility rats' tissues of gastric antrum. Methods 70 Wistar rats were randomly divided into 7 groups, 10 rats in each group,model group were irritated stomach with different doses(Sg/kg,10g/kg,15g/kg,20g/kg, 40g/kg) of liquorice decoction for 5 days,normal group were given the same volume of normal saline for 5 days. Each group rats were fasting without water deprivation in 24h before administering the medicine. Then to test the gastric electrical activity and 5 - HT APUD cell number . dimension, average grey values in low gastric motility rats' tissues of gastric antrum with immuno--histochemical method. Results Small doses of liquorice decoction could make rat model of low gastric motility, and make 5 - HT APUD cell number,dimension reduce in rat tissues of gastric antrum, at the same time make 5 - HT APUD cell average grey values increase, campared with model groups, there were all significance difference (P 〈 0.01 ). And with the increase of dose, the gastric electrical activity were improved , the 5 - HT APUD cell number,dimension were increasing in rat's tissues of gastric antrum, the 5 - HT APUD cell average grey values were reducing, but campared with model groups, there were difference (P 〈 0.05 ). Conclusion The small doses of the liquorice decoction could reduce gastrointestinal dismotility of rats by reducing 5 -HT APUD cell number,dimension, and increasing 5 -HT APUD cell average grey values.
