摘要
目的观察大黄素对卵巢癌SKOV3细胞的增殖、凋亡和凋亡抑制蛋白Survivin和X染色体连锁凋亡抑制蛋白(XIAP)表达的影响,探讨大黄素对卵巢癌细胞增殖的作用机制。方法体外培养SKOV3细胞,分对照组(DMSO组)和大黄素组(终浓度为12.5、25和50μmol/L),MTT法测定SKOV3细胞增殖率;流式细胞术检测细胞凋亡;RT-PCR和Western blot法分别检测SKOV3细胞中Survivin和XIAP mRNA和蛋白表达。结果与对照组相比,大黄素加药组明显抑制细胞增殖并促进细胞凋亡(P<0.01)。大黄素作用后XIAP、Survivin mRNA和蛋白水平较对照组降低(P<0.01)。结论大黄素通过抑制凋亡抑制蛋白Survivin和XIAP表达促进SKOV3细胞凋亡。
Objective To investigate the effects of emodin on proliferation and apoptosis of ovarian cancer SKOV3 cells and the expression of Survivin, X-linked inhibitor of apoptosis protein (MAP) and the possible mechanism. Methods SKOV3 cells were cultured and divided into control group and emodin groups (final emodin concentra- tions were 12.5, 25 and 50 μmol/L respectively). The SKOV3 cells viability was measured by MTT and the cell apoptosis was determined by flow cytometry analysis. The changes of mRNA and protein were examined by RT-PCR and western blot respectively. Results The proliferation of SKOV3 cells in each group decreased in the different concentrations of emodin as compared with control group (P 〈 0. 01 ) ; The apoptosis rate of SKOV3 cells increased in the different concentrations of emodin groups respectively compared with control group(P 〈0. 01 ). The expres- sion of Survivin and XIAP mRNAs and proteins in SKOV3 cells in the presence of different concentration of emodin decreased as compared with control group (P 〈 0. 01 ). Conclusions Emodin may induce apoptosis of ovarian caneer SKOV3 cells by down-regulation of the expressions of Survivin and XIAP proteins.
