高效液相色谱法同时测定栀子金花丸中9个成分的含量 被引量:10

Simultaneous determination of nine compounds in Zhizi Jinhua pills by HPLC wavelength switching method

摘要 目的:建立高效液相色谱法同时测定栀子金花丸中9个成分的方法。方法:采用DIKMA Platisil ODS色谱柱(250 mm×4.6 mm,5μm),柱温35℃,流动相为甲醇-0.1%磷酸水溶液,梯度洗脱(0-5 min,40%A;5-20 min,40%A→60%A;20-26 min,60%A→60%A;26-35 min,60%A→90%A;35-52 min,90%A;52-54 min,90%A→40%A),流速为1 mL·min-1,变换波长检测(327 nm,绿原酸、盐酸小檗碱;254 nm,栀子苷、黄芩苷、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)。结果:绿原酸、栀子苷、盐酸小檗碱、黄芩苷、芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚的线性范围分别为0.02-0.4μg(r=0.9998)、0.049-0.98μg(r=1.000)、0.05-1μg(r=0.9999)、0.1-2μg(r=1.000)、0.0025-0.05μg(r=0.9996)、0.001-0.02μg(r=0.9997)、0.0014-0.028μg(r=0.9998)、0.004-0.08μg(r=0.9998)、0.00033-0.0066μg(r=0.9998),平均回收率在98.8%-100.4%之间。结论:该法经方法学验证,是可用于栀子金花丸质量控制的一个有效的方法。 Objective:To establish a method for simultaneous determination of chlorogenic acid,geniposide,berberine,baicalin,aloeemodin,rhein,emodin,chrysophanol and physcion in Zhizi Jinhua pills. Methods:The separation was performed on a DIKMA Platisil ODS column( 250 mm × 4. 6 mm,5 μm) with gradient elution( 05 min,40% A; 520 min,40% A→60% A; 2026 min,60% A→60% A; 2635 min,60% A→90% A; 3552 min,90% A; 5254 min,90% A→40% A) using methanol and 0. 1% phosphoric acid aqueous solution as the mobile phase. The flow rate was 1 mL·min1and the detection wavelengths were 327 nm for chlorogenic acid and berberine,254 nm for geniposide,baicalin,aloeemodin,rhein,emodin,chrysophanol and physcion,respectively.Results:Linear ranges of chlorogenic acid,geniposide,berberine,baicalin,aloeemodin,rhein,emodin,chrysophanol and physcion concentrations were 0. 020. 4 μg( r = 0. 9998),0. 0490. 98 μg( r = 1. 000),0. 05-1 μg( r = 0. 9999),0. 12 μg( r = 1. 000),0. 00250. 05 μg( r = 0. 9996),0. 0010. 02 μg( r = 0. 9997),0. 00140. 028 μg( r = 0. 9998),0. 0040. 08 μg( r = 0. 9998),0. 000330. 0066 μg( r = 0. 9998),respectively. Their average recoveries fell between 98. 8% and 100. 4%. Conclusion:The methodology validation shows that this HPLC method established can be used to determine the contents of chlorogenic acid,geniposide,berberine,baicalin,aloeemodin,rhein,emodin,chrysophanol and physcion in Zhizi Jinhua pills. The method is an effective method for the quality control of Zhizi Jinhua pills.
