摘要
目的:探讨化痰通络汤对大脑中动脉栓塞(MCAO)模型大鼠脑组织细胞间黏附分子(ICAM-1)、基质金属蛋白酶9(MMP9)表达的影响。方法:32只SD大鼠按体质量随机分为假手术组、模型组、尼莫地平组与化痰通络组。以大脑中动脉栓塞法建立MCAO大鼠模型,观察大鼠一般状况,Zea-longa神经功能评分,免疫组织化学染色法检测大鼠脑组织ICAM-1、MMP9表达。结果:与假手术组比较,模型组大鼠神经功能评分明显增高(P<0.01),脑组织ICAM-1、MMP9表达增强(P<0.01);与模型组比较,化痰通络组大鼠神经功能评分降低(P<0.05),脑组织ICAM-1、MMP9表达明显减弱(P<0.01)。结论:化痰通络汤可明显改善MCAO模型大鼠的神经功能和一般症状,其机制可能与抑制大鼠ICAM-1神经炎症标志物、MMP9脑组织损伤标志物的表达有关。
Objective: To investigate the influence of Huatan Tongluo Decoction on expression of ICAM-1 and MMP9 in rats brain with middle cerebral artery occlusion (MCAO). Methods: Thirty-two SD rats were randomly divided into sham operation group (sham group), model group, Nimodipine (NMDP) group and Huatan Tongluo Decoction (HT) group. The rat MCAO model was established with middle cerebral artery occlusion method. The changes of ordinary condition were observed and the Zea-longa neurology scores were measured. The expressions of ICAM-I and MMP9 in rats' brain were detected with immunohistochemistry. Results: The Zea-longa neurological scores in model group were significantly higher than that in the sham group (P〈0.01), as well the expressions of ICAM-1 and MMP9 in brain tissues (P〈0.01). The Zea-longa neurological scores in HT group were lower than that in the model group (P〈0.05), as well the expressions of ICAM-1 and MMP9 (P〈0.01). Conclusion: Huatan Tongluo Decoction could significantly improve the neurological function and ordinary condition of MCAO rats, the mechanism of which might be related to inhibiting the expression of the neuro-inflammatory biomarker ICAM-1 and the marker of tissue injury MMP9.
