摘要
目的 建立枇杷叶总三萜酸固体分散体片中6种三萜酸含量测定方法。方法 采用HPLC-ELSD,色谱条件为色谱柱Ultimate XB C18柱(4.6 mm×250 mm,5μm),流动相乙腈-甲醇-0.5%醋酸铵溶液,梯度洗脱,流速1.0 mL·min-1,柱温35℃,蒸发光散射检测器漂移管温度100℃,载气流速2.5 L·min-1。结果 6种三萜酸蔷薇酸、委陵菜酸、马斯里酸、科罗索酸、齐墩果酸、熊果酸分别在0.583~5.832,0.366~3.664,0.335~3.352,0.533~5.326,0.257~2.570,0.602~6.018μg内峰面积积分值常用对数与进样量常用对数有良好的线性关系;平均回收率分别为101.83%、100.63%、103.42%、103.59%、104.07%、104.56%,RSD分别为1.37%、2.77%、2.03%、1.63%、1.42%、1.84%。结论 本实验所建立的含量测定方法简便、准确、分离效果好、重复性强,可用于枇杷叶总三萜酸固体分散体片的质量控制。
OBJECTIVE To establish a method for determination of six triterpene acids in solid dispersion tablets of Eriobotryae Folium total triterpenoid acids by HPLC-ELSD. METHODS The samples were analyzed with Ultimate XB Cj8 column ( 4. 6 mm× 250 mm,5 μm) and eluted with acetonitrile-methanol-0. 5% ammonium acetate solution as mobile phases in gradient mode, the flow rate was 1.0 mL · min-1 , the column temperature was 35 ℃, the drift tube temperature of ELSD was set at 100 ℃ ,and the flow rate of nitrogen was 2. 5 L · min - i. RESULTS There were good linear relationship between natural logarithm of peak areas and injection quality in the range of 0. 583 - 5. 832, 0. 366 - 3. 664, 0. 335 - 3. 352, 0. 533 - 5. 326, 0. 257 - 2. 570, and 0. 602 - 6. 018 μg for rosolie acid, tormentic acid, maslinic acid, corosolic acid, oleanolic acid, and ursolic acid, respectively. The average recoveries of these six triterpene acids were 101.83% ( RSD 1.37% ) , 100. 63% ( RSD 2.77% ), 103.42% ( RSD 2. 03% ), 103.59% ( RSD 1.63% ) , 104. 07% ( RSD 1.42% ), and 104. 56% ( RSD 1, 84% ), respectively. CONCLUSION The established method is sim- ple, accurate and repeatable with good resolution, which can be used for the quality control of solid dispersion tablets of Eriobotryae Folium total triterpenoid acids.
