rhGM-CSF、纳米银对深Ⅱ°烫伤愈合过程炎症反应的影响 被引量:5

Inhibition of Recombinant Human Granulocyte/Macrophage Colony Stimulating Factor and Nano-Silver on the Inflammation of Deep Burn DegreeⅡ

摘要 目的:研究重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)、纳米银对深Ⅱ°烫伤创面愈合过程炎症反应的影响。方法:用Wistar大鼠建立深Ⅱ°烫伤模型,分为A、B、C 3组,A组(30例):凡士林纱布覆盖;B组(30例):纳米银覆盖;C组(30例):rhGM-CSF涂抹创面。伤后第1,4,7,10,14,21天,观察创面炎症反应,细菌培养并计数,按照酶联免疫吸附法测定血清中白细胞介素-2(IL-2)、白细胞介素-8(IL-8)水平。结果:炎症反应,A组>B组>C组;A组、B组/C组分别于伤后第4、10天出现细菌生长,B组、C组与A组相比,细菌生长数量少,并且第10,14,21天差异有统计学意义(P<0.05),B组和C组之间差异无统计学意义(P>0.05);IL-2水平,伤后第1天各组间及第4天B组和C组之间差异无统计学意义(P>0.05),其余各时相点各组间差异有统计学意义(P<0.05),IL-8水平,伤后第1天,A组和B组、B组和C组之间无统计学意义(P>0.05),其余各时相点各组间差异均有统计学意义(P<0.05)。结论:rhGM-CSF、纳米银外用均能减少创面细菌生长,减轻炎症反应,并且rhGM-CSF在减轻创面炎症反应方面,更有优势。 Objective:To study the effect of recombinant human granulocyte/macrophage colony stimulating factor(rhGM-CSF)and nano-silver on deep burn degreeⅡ through observing the changes of inflammatory reaction during the healing process.Methods:The burn models were established on Wistar rats,and then were randomly divided into three groups according to petrolatum treatment(group A,n=30),nano-silver treatment(group B,n=30),and rhGM-CSF treatment(group C,n=30),respectively.Bacteria samples were extracted from the wound for culturing and counting after 1,4,7,10,14,and 21day(s)respectively of treatment.The serum concentrations of interleukin-2(IL-2)and interleukin-8(IL-8)were measured in serum with ELISA.Results:The most severe inflammatory reaction was found in group A,while the mildest one wasfound in group C.Bacteria were found from the 4th day in group A and from the 10 th day in group B and group C after treatment,and the bacterial counting was less in group B and group C than in group A on the 10 th,14th,and 21 st days(P〈0.05),but there were no significant difference between group B and group C in bacterial counting(P〉0.05).Except on the 1st or4 th days,significant difference was found among different groups and different time points in serum IL-2and IL-8(P〈0.05).Conclusion:rhGM-CSF and nano-silver could alleviate the inflammatory reaction after burn and inhibit the bacterial growth,but rhGM-CSF showed better effect than nano-silver.
