高压脉冲电场协同酶法提取竹叶中活性成分的研究 被引量:7

摘要 采用高压脉冲电场协同酶法技术从竹叶中综合提取竹叶黄酮、茶多酚和竹叶多糖三种活性成分。通过正交试验确定酶法水提多糖、高压脉冲电场技术提取竹叶黄酮和茶多酚的最优化工艺。结果在复合酶浓度为0.8%,温度55℃,pH 5.0,酶解80 min的条件下,竹叶多糖提取率为3.47%,提取多糖后的滤渣与60%乙醇按料液比1∶20匀质,在电场强度22 kV/cm,脉冲数为9的条件下进行高压脉冲电场提取,黄酮提取率达到2.49%,茶多酚提取率可达1.26%。该工艺可有效地从竹叶中联合提取黄酮、茶多酚和多糖。 Enzyme combined with pulse electric field technology was used to extract three active ingredients from bamboo leaves.The three ingredients include flavonoids,polyphenols and polysaccharides.Orthogonal experiment showed that the enzyme combined with pulse electric field extraction was the best in extracting the active ingredients.The best extract conditions were:enzyme concentration 0.8%,extraction temperature 55℃,pH value 5.0,the enzyme hydrolysis 80 min.The extraction rate of polysaccharides of bamboo leaves was 3.47%,after polysaccharides extraction,the residue was extracted with 60% ethanol at solid-liquid ratio of 1 ∶ 20,the electric field intensity 22 kV/cm pulse and frequency of 9.Under the above parameters,the extraction rate for flavonoids and polyphenols were 2.49 % and 1.26 %,respectively.The conclusion is:enzymatic combined with pulse electric field technology is an effective method to extract flavonoids,tea polyphenols and polysaccharides from bamboo leaves.
