摘要
目的通过观察6周不同强度跑台运动和白藜芦醇给药,对非酒精性脂肪肝大鼠肝组织Bax,Bcl-2指标变化及凋亡肝细胞数目的影响,从肝细胞凋亡的角度探讨运动和白藜芦醇给药对非酒精性脂肪肝大鼠改善的可能机制。方法将SPF级雄性SD大鼠70只,分为普通安静对照组(NC)、非酒精性脂肪肝模型组(HF)、非酒精性脂肪肝小强度运动组(LE)、非酒精性脂肪肝中等强度运动组(ME)、非酒精性脂肪肝递增强度运动组(IE)、白藜芦醇组(RN)、白藜芦醇中等强度运动组(RE)。共计7组,每组10只大鼠。各运动组每次运动时间60 min,每周运动5 d。白藜芦醇灌胃每周7 d,共计6周。免疫组织化学法测试Bax、Bcl-2的表达,用TUNEL方法测试肝细胞凋亡情况。结果 (1)6周跑台运动和白藜芦醇给药干预有效降低Bax的表达,运动组中中等强度表达量最低(P<0.01),联合干预组较运动组显著下降(P<0.05),中等强度运动组Bcl-2升高(P<0.01),联合干预组较中等强度运动组升高(P<0.05);(2)6周跑台运动和白藜芦醇给药干预有效降低肝细胞凋亡的表达(P<0.05),联合干预组较其中等强度运动组显著降低了肝细胞凋亡(P<0.05)。结论实验通过6周的跑台运动和白藜芦醇单独或联合的干预方式,Bax表达下降,而Bcl-2表达增加,肝细胞的凋亡数量减少,说明两种干预方式均有效的保护肝脏,有促进NAFLD的转归的作用,而联合干预方式具有转归的叠加效应。
Objective Observing the effects of 6 - week treadmill exercise with different intensity and resveratrol on Bax, Bcl - 2 and liver cell apoptosis of nonalcoholic fatty liver disease rats to discuss the improving effects of exercise and resveratrol on nonalcoholic fatty liver disease rats. Methods 70 SPF male SD rats were divided into the control group ( NC ) , model group of nonalcoholic fatty liver disease (HF) , low intensity exercise group of nonalcoholic fatty liver disease (LE) , moder- ate intensity exercise group of nonalcoholic fatty liver disease (ME) , increment intensity exercise group of nonalcoholic fatty liver disease (IE) , resveratrol group (RN) , resveratrol with moderate intensity exercise group(RE). There were 10 rats in each group. Exercise was arranged to 60 min per day and 5 days per week. Resveratrol was given a gavage as 7 days a week. All the intervention lasted for 6 weeks. The expression of Bax and Bcl - 2 were determined by immune histochemical method, and test liver cell apoptosis was determined by TUNEL method. Results ( 1 ) 6 - week treadmill exercise and resveratrol ef- fectively reduced the expression of Bax in rats, intervention group of moderate intensity was the best in the three exercise intervention groups( P 〈 0. 01 ), joint intervention group was the best of all( P 〈 0.05 ). Instead, The expression of Bcl - 2 in rats of each intervention group increased( P 〈 0.01 ) and that of the joint intervention group was the highest in all intervention groups ( P 〈 0.05 ) ; ( 2 ) 6 - week treadmill exercise and/or resveratrol intervention effectively reduced the expression of liver cell apoptosis ( P 〈 0.05 ), and the joint intervention group significantly decreased than other groups ( P 〈 0. 05 ). Conelusions In this study, the expressions of Ban, and the number of the apoptosis of liver cells decreased after six - week tread- mill exercise and/or resveratrol, while the expression of Bcl - 2 increased, which showed that six - weeks treadmill exercise and/or resveratrol could were effective in protecting the liver and had the effect of ameliorating NAFLD, the joint intervention with a superimposed effect in all intervention groups.
