糖尿病动物模型视网膜病变的实验研究

摘要 目的:建立糖尿病性视网膜病变(DR)动物模型。方法:叙利亚金黄地鼠20只随机分为正常对照组和模型组,采用腹腔内注射2%链脲佐菌素造模,观察两组动物一般状态、血糖、眼底彩照、荧光素眼底血管造影及光学相干断层扫描的区别。结果:模型组出现明显的"三多一少"糖尿病临床表现,血糖值明显高于正常对照组(P<0.01),眼底检查可见黄白色点状病灶及小片状红色病灶,FFA检查出现视网膜平均循环时间延迟和视网膜点状及小片状高荧光灶的改变,OCT检查可见神经上皮层间的颗粒样强反射。结论:2%链脲佐菌素致叙利亚金黄地鼠高血糖模型,与人类糖尿病及糖尿病性视网膜病变的表现十分相似,该模型可用于研究DR的发生、发展机制。 Objective:To establish animal models of diabetic retinopathy. Method: 20 Syria golden hamsters were randomly divided into normal control group and model group.Diabetes animal models were induced by intraperitoneal injection of 2% streptozotoein. The difference in the general condition,blood glucose,fundus photograph,fundus fluoreseein angiography (FFA)and optical coherence tomography (OCT)of the animals were observed between two groups. Result:The model group animals appeared obviously clinical manifestations of diabetes, such as polydipsia,polyphagia,polyuria and weight loss. Blood glucose was significantly higher than that of normal control group(P〈0.01 ). Fundus examination showed yellow white dot lesions and small pieces of red lesions. FFA test emerged average retinal circulation time delay and retinal dot and flake high fluorescent focus. OCT examination expressed graininess strong reflection in neuroepithelial layer. Conclusion: Performances of Syria golden hamsters diabetes model induced by 2% streptozotocin is similar to those of human diabetes and diabetic retinopathy, so the models can be used to study occurrence and development mechanism of diabetic rctinopathy.
