解磷钾胶质芽孢杆菌培养基的优化研究 被引量:1

摘要 通过优化单因子及正交实验对影响胶质芽孢杆菌生物量的主要因子碳源、氮源的种类及添加量,分析出菌体生物量的最大影响因子和培养基组分的最优组合。结果表明:由单因子实验可知以葡萄糖为碳源,硫酸铵为氮源时菌体生物量较高。正交实验结果可知葡萄糖是菌体生物量主要的影响因子;比较理想的培养基优化配方为(g/L):葡萄糖24,硫酸铵1,豆饼9,碳酸钙2,硫酸镁2,磷酸二氢钾4,氯化铁0.1。在此条件下活菌数可达8×108cfu/mL。 The main influence factors,including carbon sources,nitrogen source,which influence the bacterial growth,was optimized through single factor; Orthogonal experiment was used to optimize the additive quality of carbon sources and nitrogen source,and analyze the maximum impact factor of the bacterial biomass and the optimal combination of culture media components.The results showed that:by single factor experiment we can know there is higher bacteria biomass when glucose was used for carbon source,and ammonium sulfate was used for nitrogen source.The results of orthogonal experiment showed that glucose was the main impact factors of the bacterial biomass; the optimization of the ideal culture medium formula is (g/L):glucose 24,ammonium sulfate 1,DouBing 9,calcium carbonate 2,magnesium sulfate 2,ammonium sulfate 2,potassium dihydrogen phosphate 4,ferric chloride 0.1.Under this condition,the living bacterium number can reach 8 × 10s cfu/mL.
