MACC1在人食管鳞癌中的表达及对侵袭的影响 被引量:2

Expression of MACC1 in human esophageal squamous cell carcinoma and its function in metastasis

摘要 目的分析结肠癌转移相关基因1(metastasis-associated in colon cancer-1,MACC1)在人食管鳞癌组织中的表达及其与临床病例特征间的关系;研究MACC1基因在食管鳞癌侵袭转移中的生物学功能。方法采用免疫组化S-P法检测MACC1在食管鳞癌组织中的表达,结合临床病理特征分析MACC1表达与食管鳞癌侵袭、迁移之间的关系及其临床意义;采用脂质体法将质粒shRNA-MACC1转染食管鳞癌细胞株ECA109,采用G418压力培养筛选稳定转染ECA109阳性细胞,通过荧光定量PCR、Western blot检测ECA109阳性细胞MACC1基因和蛋白的表达,通过MTT、Transwell检测ECA109阳性细胞增殖能力、侵袭能力的变化。结果免疫组化结果显示食管鳞癌组织MACC1阳性率(94.1%)显著高于癌旁组织(37.3%),差异具有统计学意义(P<0.05);食管鳞癌上皮细胞中MACC1强阳性率(90.0%)显著高于非典型增生食管上皮细胞(15.0%),差异具有统计学意义(P<0.05),未明显增生食管上皮细胞中MACC1表达为阴性;在性别、年龄、肿瘤大小、病变部位、分化程度以及浸润深度分组比较中,MACC1蛋白表达差异无统计学意义(P>0.05)。在体外实验中,随着MACC1表达下调,ECA109细胞增殖能力、侵袭迁移能力明显下降。结论 MACC1与食管鳞癌侵袭迁移密切相关;MACC1在非典型增生食管上皮和食管鳞癌组织中异常表达,在由异常增生到食管鳞癌的过程中,MACC1蛋白表达随之升高,提示MACC1基因与食管鳞癌发生、发展密切相关。 Objective To determine the expression of metastasis-associated in colon cancer-1( MACC1) in human esophageal squamous cell carcinoma( ESCC),and investigate its correlation with clinical features for its role in the invasion and metastasis of the tumor. Methods The expression of MACC1 in 51 ESCC specimens and para-carcinoma was measured by immunohistochemical staining. The correlation of the MACC1 expression with the clinicopathological features was analyzed in the invasion and metastasis of ESCC.The plasmid harboring MACC1-shRNA was transfected into ESCC cell line ECA109. The stable cells were screened out by G418 and identified by real-time PCR and Western blotting. Then the proliferation,migration and invasion of the positive ECA109 cells were studied by MTT assay and Transwell chamber test. Results The positive rate of MACC1 was 94. 1% in the obtained ESCC tissues,and was only 37. 3% in the corresponding para-cancer tissues( P〈 0.05). MACC1 was strongly expressed in the epithelial cells of ESCC( 90. 0%),significantly higher than it in the epithelial cells of atypical hyperplasia esophagus( 15. 0%,P〈 0.05). No MACC1 expression was found in the epithelial cells of esophagus without obvious hyperplasia. There was no statistically significance in the gender,age,tumor size,location,differentiation and depth of invasion among the ESCC with different MACC1 levels( P〉 0.05). In the in vitro study,MACC1-shRNA transfection resulted in significantly reduced proliferation,migration and invasion in the ECA109 cells. Conclusion MACC1 is closely involved in the invasion and metastasis of ESCC. It is abnormally expressed in the esophagus epithelium with atypical hyperplasia and in the ESCC. In the process of atypical hyperplasia to carcinoma,the protein expression of MACC1 is elevated,suggesting its intimate role in the incidence and development of ESCC.
