贝母素甲、贝母素乙对4T1乳腺癌细胞炎性微环境的干预调节作用 被引量:16

摘要 目的:研究贝母素甲、乙对4T1乳腺癌细胞的干预抑制及对其肿瘤炎性微环境的调节作用。方法:CCK-8检测贝母素甲、乙对4T1细胞抑制率;ELISA法检测炎性相关因子TGF-β、VEGF、MMP-9、MCP-1的分泌量;RT-PCR检测TGF-β、VEGF、MMP-9mRNA表达情况。结果:贝母素甲、乙对4T1乳腺癌细胞具有抑制作用;贝母素甲、乙可分别下调4T1细胞TGF-β,VEGF、MCP-1及MMP-9分泌量(P<0.05,P<0.01);贝母素甲、乙可降低4T1细胞TGF-β、VEGF等mRNA表达(P<0.05,P<0.01)。结论:贝母素甲、乙对4T1乳腺癌细胞具有显著抑制作用,可通过抑制炎性相关因子的释放调节微环境。 Objective: To study the effects of Peimine and Peiminine on intervening 4T1 cells and regulating its inflammatory microenvironment. Methods: Breast cancer 4T1 cells were intervened by Peimine and Peiminine with different concentrations in vitro at 24 h, 48h and 72 h. The inhibition ratios of 4T1 ceils were measured by CCK-8 assay at different times. Secretion of inflammation associated factors TGF-β, VEGF, MMP-9 and MCP-1 was detected through ELISA. The mRNA expressions of TGF-[3, VEGF and MMP-9 were assessed by RT-PCR. Results: Peimine and Peiminine within 5 μM/L to 100 μM/L intervened 4TI cells to show the inhibition effect on gradient increasing trend. The secretion of TGF- 13, VEGF and MCP-1 was inhibited by Peimine significantly (P 〈 0. 05, P 〈 0. 01 ). Peiminine inhibited TGF-β, MMP-9 and MCP-1 secretion (P 〈 0. 05, P 〈 0.01 ). The mRNA expression of TGF-β and VEGF was suppressed by Peimine and Peiminine in 4T1 (P 〈 0. 05, P 〈 0. 01 ) , and Peiminine could down regulated the expression of MMP-9 ( P 〈 0.05 ). Conclusions: Peimine and Peiminine had distinct inhibiting effect on 4T1 ceils, and regulated its microenvironment through controlling the release of inflammatory factors.
