摘要
为更好地了解近年来仔猪流行性腹泻疫情,收集上海地区猪流行性腹泻病猪的临床样品,扩增猪流行性腹泻病毒保护性抗原基因(COE基因),构建原核表达载体pColdⅡ-COE,转化至大肠埃希菌pGTf2/BL21,IPTG诱导表达,SDS-PAGE结果显示,表达的COE蛋白大小为16ku,Western blot分析表明,该蛋白与临床上猪流行性腹泻病毒阳性猪的血清有特异性反应,说明所表达的COE蛋白具有反应原性,为进一步研制猪流行性腹泻疫苗和ELISA检测方法奠定了基础。
In order to understand recent outbreaks of porcine epidemic diarrhea (PED),we collected a few clinical samples from swine with PED in Shanghai .COE gene was amplified and cloned into prokaryotic expression vector pColdⅡ,to construct a recombinant expression plasmid pColdⅡ-COE.And then the pColdⅡ-COE was transformed into Chaperone competent cell pG-Tf2/BL21.The expressed products were analyzed by SDS-PAGE and Western blotting.Results showed that COE protein was expressed with mo-lecular mass of 16ku and could specifically reacted with PEDV positive serum.Our results proved the COE protein had good immunogenicity and could provided a basis for the study of vaccine and ELISA of PEDV.
