龙牡壮骨颗粒治疗大鼠佝偻病药效学及其机制 被引量:7

Effects and mechanism of actions of Longmu Zhuanggu granules in rickets rats

摘要 目的:研究中药复方龙牡壮骨颗粒对维生素D缺乏佝偻病的药理作用及作用机制。方法:将30只雄性SD大鼠随机均分为空白组,模型组和龙牡壮骨颗粒组,模型组和龙牡壮骨颗粒组用维生素D缺乏饲料喂养并避光处理4周诱导维生素D缺乏性佝偻病,之后给予正常饲料。空白组和模型组给予0.9%生理盐水、龙牡壮骨颗粒组给予龙牡壮骨颗粒水剂,灌胃给药,每天一次,连续给药8周。期间每周检测大鼠体质量变化,给药结束后处死大鼠,检测身长、骨形成、代谢、血生化指标以及钙跨膜转运相关蛋白变化。结果:龙牡壮骨颗粒能显著增加佝偻病大鼠体质量和身长(P<0.05);同时提高骨干重、骨灰重、骨密度、血磷、25-羟基维生素D3[25(OH)D3]含量及显著降低碱性磷酸酶(ALP)及抗酒石酸酸性磷酸酶(TRAP)含量(P<0.05),其中骨钙含量和血钙含量增加尤其明显(P<0.01);深入研究发现其能显著增加钙转运蛋白(CAT1)和维生素D受体(VDR)蛋白表达量,并能一定程度地提高质膜钙泵(ATP2B1)蛋白的表达。结论:龙牡壮骨颗粒能显著增加佝偻病大鼠对钙的吸收及跨膜转运能力,并能促进小肠对维生素D的摄取,增加佝偻病大鼠骨钙、骨盐含量,发挥治疗佝偻病作用。 OBJECTIVE To investigate the therapeutic effects and the mechanisms of actions of Longmu Zhuanggu granules in rickets rats.METHODS Thirty SD rats were randomly divided into three groups,namely control group,model group and Longmu Zhuanggu granules group.The mice of model group and Longmu Zhuanggu granules group were fed with vitamin D deficiency diet and shielded from light for 4 weeks to induce rickets.After being administrated with Longmu Zhuanggu granules by gavage every day for 8 weeks,the body weight and length of rats were measured every week.After mice were sacrificed,the dry weight,density and calcium content of the femur were detected to confirm the effects of Longmu Zhuanggu granules on bone formation and resorption.Blood biochemical indicators were detected too.To investigate the potential mechanisms,expression of several calcium transport related proteins in small intestine were tested.RESULTS Longmu Zhuanggu granules was effective on increasing the body weight and length of rickets rats(*P〈0.05),and could elevate bone dry weight,bone ash weight,bone mineral density,blood phosphate,25(OH)D3and it also could lower the level of ALP and TRAP in the blood significantly(P〈0.05),while the bone calcium and blood calcium were increased(P〈0.01).It was found that Longmu Zhuanggu granules could obviously up-regulate the expression of CAT1,VDR and ATP2B1.CONCLUSION Longmu Zhuanggu granules can elevate bone calcium and mineral content by augmenting calcium absorption,calcium transport and promoting the intake of vitamin D in the small intestine,which may play critical roles on rickets treatment.
