摘要
目的通过分析电针对慢性情绪应激(CES)焦虑大鼠中枢5-羟色胺(5-HT)水平的调节作用及对其代谢产物含量的影响,探讨防治焦虑症发病的部分神经生化机制。方法采用不可预知的情绪应激刺激方法,刺激大鼠21d,建立CES焦虑模型。电针"百会"、"三阴交"穴治疗21d,采用高架十字迷宫测试系统观测焦虑情绪的行为学变化,运用荧光分光光度法测定大鼠下丘脑、杏仁核区单胺神经递质5-HT及其代谢产物5-羟吲哚乙酸(5-HIAA)的含量。结果模型组较空白组大鼠5-HT及5-HIAA含量明显升高(P<0.05,或P<0.01);电针可降低模型大鼠下丘脑、杏仁核区单胺神经递质5-HT及5-HIAA的含量。(P<0.05或P<0.01)。结论电针抗焦虑作用可能与降低下丘脑和杏仁核区5-HT含量及调节其代谢产物含量密切相关。
Objective To research the regulation effect of electro-acupuncture on the levels of central 5-HT and its metabolites in rat with chronic emotional stress-induced anxiety,and discuss the neural biochemical mechanism of the anxiety attack..Methods Chronic emotional stress anxiety model was established by using chronic unpredictable emotional stress methods for 21 days. Electric acupuncture was applied to Baihui(GV20) and Sanyinjiao(SP6) and lasted for 21 days. Observating anxiety behavior change with the Elevated Plus Maze test system. The contents of 5-HT and its metabolite 5-HIAA in the hypothalamus and amygdala region were measured by fluorescence spectrophotometry. Results Compared with control group,the contents of 5-HT and its metabolite 5-HIAA in the hypothalamus and amygdala region increased obviously in model group(P〈0. 05,or P〈0.01). Electric acupuncture can decrease the contents of 5-HT and its metabolite 5-HIAA of the model group in the hypothalamus and amygdala region(P〈0. 05 or P〈0. 05). Conclusion Anti-anxiety effect of electric acupuncture is possibly closely related to reduce the content of 5-HT and its metabolite 5-HIAA in the hypothalamus and amygdala region.
