A2菌株发酵液中抑菌成分的分离及结构鉴定

摘要 [目的]采用分离手段鉴定A2菌株发酵液中抗菌成分。[方法]采用阳离子交换树脂、CM-Sephadex-C25凝胶层析、离子对液相色谱制备等分离手段对A2菌株发酵液进行分离,通过核磁、质谱对化合物进行结构表征。[结果]分离得到3个化合物(H2、H3、H4),采用抑制孢子萌发速率法和抑制菌丝生长法对3个化合物进行抑菌活性测定,实验结果表明3个化合物对真菌和细菌均有较好的抑制作用。[结论]通过核磁、质谱分析,参考相关文献,化合物H2、H4经结构鉴定分别为链丝菌素D和链丝菌素F。 [Aims] The aim of this investigation is to identify the antimicrobial ingredients in the metabolites of strain A2. [Methods] After extracted with cation resin, separated by CM-Sephadex-C25 gel chromatography and ion-pair reversed-phase HPLC, the obtained active ingredients were finally identified by the techniques of NMR spectroscopy and mass spectrometry. [Results] Three compounds(H2, H3, H4) were obtained, and their antifungal and antibacterial activities were evaluated by spore germination and agar diffusion methods, respectively. The results indicated that all compounds exhibited strong antifungal and antibacterial activity. [Conclusions] Base on the data of NMR and MS, and by comparing with related literatures, H2, H4 were identified as streptothricin D and streptothricin F, respectively.
