摘要
为了优化文心兰生根培养基,以文心兰不定芽为材料,1/2 MS为基本培养基,在(25±1)℃,30μmol·(m2·s)-1,10h·d-1光照时间的条件下优化文心兰生根配方,同时筛选移栽基质。结果表明:IBA是影响文心兰不定芽生根的主导因子,其次是蔗糖;1/2 MS+IBA 0.7mg·L-1+NAA 0.4mg·L-1+蔗糖40g·L-1有利于文心兰不定芽的生根,生根率98.49%、根数9.1条、根长1.24cm、茎高1.49cm、苗高5.08cm、茎粗0.30cm;刨花是文心兰较佳的移栽基质,整体效果高于其它基质,60d后苗高6.75cm,茎粗0.55cm,鲜重1.20g,根冠比0.44。
In order to optimize rooting medium for Oncidium,taking adventitious bud of Oncidiumas materials and 1/2 MS as the basic culture medium,rooting medium formula was optimized and transplanting substrate was screened under(25±1)℃,illumination intensity of 30μmol(m2·s)-1 and 10 hlight every day.The results showed that IBA was the dominant factor influencing Oncidiumadventitious bud rooting,next was the sucrose.The medium of 1/2 MS+0.7mg·L-1 IBA+0.4mg·L-1 NAA+40g·L-1 sugar+5g·L-1 had the best benefit for rooting,the rooting rate was 98.49%,the rooting number was 9.1branches,the root length was 1.24 cm,seedling height was 5.08 cm,stem diameter was 0.30 cm.Wood shaving was better transplanting substrate,the overall effect was obviously better than other substrates,seedling height was 6.75 cm,stem diameter was0.55 cm,root-shoot ratio was 0.44 after 60d.
